摘 要:目的探讨扶正解毒颗粒(FJK)对镍致大鼠睾丸生殖细胞毒作用的拮抗作用。方法选择健康成年Wistar雄性大鼠,硫酸镍(NiSO4)2.5mg/kg腹腔注射染毒,FJK2.5、5.0和10.0灌胃处理。采用流式细胞术检测各组大鼠睾丸组织中各倍体细胞所占的比率及其细胞周期的变化;透射电子显微镜观察睾丸细胞的形态学变化。结果NiSO4可使睾丸组织G0/G1期细胞数减少,G2/M期细胞数增加(P<0.05);各倍体细胞比率的分析发现,1C∶2C、4C∶2C和4C∶S比值升高(P<0.05);用FJK处理后,FJK5.0、10.0g/kg组睾丸G0/G1期细胞数回升,G2/M期细胞数减少,各倍体细胞比率趋于正常,与NiSO4组比较差异有统计学意义(P<0.05)。电子显微镜观察显示,NiSO4可引起大鼠生殖细胞核固缩、核质间隙增宽,核膜皱缩、染色质聚集、胞浆可见空泡、线粒体肿胀等不同程度的形态学改变;用FJK处理后,FJK5.0、10.0g/kg组上述表现明显改善,尤其FJK10.0g/kg组超微结构基本恢复正常。结论FJK对NiSO4所致大鼠睾丸各倍体细胞比率、细胞周期和超微结构的异常改变均具有逆转作用。Objective To explore the antagonistic action of FuZhengJieDu granules (FJK) on the toxic effects of testicular germ cells induced by nickel sulfate( NiSO4 ). Methods The male Wister mrs were divided into five groups, including the control group with normal saline solution, NiSO4 2.5 mg/kg model group administrated intraperitoneally for 30 d, and FJK treated groups at doses of 2.5, 5.0 and 10.0 g/kg with peroral route respectively for 21 d after the rats were treated with NiSO4 2.5 mg/kg for 9 d. The various testicular cell ratios and the cell cycle were measured by Flow cytometry (FCM). The ultrastructure changes of testis cells were observed with transmission electron microscope (TEM). Results Compared with the control group, the 1 C:2C, 4C:2C and 4C : S-phase ratios of testis cells increased significantly in NISO42.50 mg/kg group( P 〈 0.05). The cells of G0/G1 and S-phase decreased, but G2/M cells increased in NiSO4 2.50 mg/kg group( P 〈 0.05). The TEM result showed that the ultrastructure of all kinds of testicular germ cells showed various abnormal changes in NiSO4 groups such as karyopycnosis, broadening of nucleoplasmic space, shrinking of nuclear membrane, chromatin aggregation, intracytoplasmic vacuolar degeneration, mitochondrial swelling, increasing of lysosome number and alveoli expansion of smooth endoplasmic reticulum and so on. After the treatment with FJK, the 1 C:2C, 4C:2C and 4C:S-phase ratios decreased and the ceils of G0/Gi and S-phase increased, meanwhile the G2/M cells decreased in FJK 5.0,10.0 g/kg groups compared with the NiSO4 exposure group( P 〈 0.05 ). The ultrastructure abnormal changes of the testicular germ cells were obviously recovered in FJK 5.0,10.0 g/kg groups.Conclusion The results showed that FJK could antagonize distinctly the abnormal changes of the cell cycle, ratios of various testis cells and ultrastructure induced by nickel sulfate.
