摘 要:研究了蒲公英总黄酮的提取条件及其抑菌性能,考查了乙醇质量分数、料液比、提取温度、提取时间和提取次数等因素对蒲公英总黄酮提取效果的影响,并在单因素试验基础上进行了正交实验以确定最佳的提取条件。试验结果表明,蒲公英总黄酮的最佳提取条件为:以40%乙醇为提取剂,在70℃、料液比1∶20条件下提取4次,每次提取时间30min,蒲公英粗总黄酮的提取率为17.51%(以芦丁计)。最佳提取条件下,相对标准偏差为0.41%,说明试验重复性很好。利用蒲公英总黄酮对黑曲霉、宛氏拟青霉、酿酒酵母和枯草杆菌进行了抑菌试验研究。结果表明,0.43g/L(以芦丁计)的蒲公英总黄酮对宛式拟青霉和枯草杆菌具有良好的抑菌作用,对酿酒酵母具有一定的抑菌作用,而对黑曲霉没有抑菌作用。An experiment was conducted to study the extraction conditions of total flavonoids from Taraxacum mongolicum and its bacteriostatic performance. The effects of ethanol volume fraction, ratio of material to liquid, temperature, extracting time and extracting times on extraction effect of total flavonoids from T. mongolicum were discussed. The optimum extraction conditions of total flavonoids by using orthogonal experiment based on single factor were obtained as ethanol solvent 40% , ratio of material to liquid 1 : 20, extraction at 70 degree C for 4 times and 30min for each time. Yield of rude total flavonoids is 17.51% (as rutin). Relative standard deviation is 0.41% under the optimum conditions, which indicates a good repeatability. Bacteriostatic performances against Aspergillus niger, Paceliomyces variotii bainier , Saccharomyces cerevisiae and Bacillus subtilis for total flavonoids from T. mongolicum were measured respectively. Results show that the total flavonoids have stronger inhibitions on P. variotii bainier and B. subtilis, and inhibit S. cerevisiae to some extent, but have no inhibition on B. subtilis under a concentration of 0.43 g/L.
