摘  要:目的:观察巴戟天多糖对成骨细胞核心结合因子α1(Cbfα1)表达的影响,并与巴戟天水提物相对照,阐述巴戟天补肾阳,强筋骨作用的实质。方法:取24h内新生SD大鼠头盖骨成骨细胞,用巴戟天多糖及巴戟天水提物药物血清进行体外培养,72h后提取成骨细胞总RNA进行RT-PCR检测核心结合因子α1mRNA相对表达量。结果:巴戟天水提物组(1.3582±0.0966)与巴戟天多糖组(1.9073±0.1280)Cbfα1mRNA与对照组(0.7347±0.0327)相比具有非常显著差异,且巴戟天多糖组非常显著优于巴戟天水提物组。结论:巴戟天水提物与巴戟天多糖均能显著提高成骨细胞Cbfα1mRNA的表达,且巴戟天多糖的这种作用优于巴戟天水提物。提示巴戟天多糖是巴戟天提高成骨细胞(OB)活性的有效成分之一。Objective: To investigate the effects of Radix M orindae Officinalis polysaccharide on expression of Cbfαt1mRNA in osteoblast, and compared to its aqueous extract. Methods: Osteoblasts were separated from the parietal bone of 24- hours old SD rat, then cultured with Radix Morindae Officinalis polysaccharide or aqueous extract of Radix Morindae Officinalis in vitro. 72 hours later, osteoblast Cbfα1 mRNA levels were measured by RTPCR method. Results: Osteoblasts Cbfα1 mRNA level in Radix Morindae Officinalis polysaecharide group or aqueous extract of Radix Morindae Officinalis group are higher than those in control group. The expression of Osteoblasts Cbfα1 mRNA in Radix Morindae Officinalis polysaccharide group is obviously higher than that in aqueous extract group. Conclusion: Both Radix Morindae Officinalis polysaccharide and aqueous extract of Radix Morindae Offici- nalis can increase the expression of osteoblast Cbfα1mRNA and the effect of Radix Morindae Officinalis polysaccharide is better than extract of Radix Morindae Officinalis. Which proved that Radix Morindae Officinalis polysaccharide is one of the active components to increase the activity of osteoblasts.
