摘  要:目的探讨Na^+-H^+交换体1(Na^+-H^+exchanger1,NHE1)反义基因磁性纳米微粒对SGC-7901胃癌细胞形态学的影响。方法构建NHE1反义基因真核表达载体和NHE1反义基因磁性纳米微粒,将NHE1反义基因转染至SGC-7901胃癌细胞中,从光镜、电镜水平观察转染与未转染细胞形态学的变化。结果氧化铁磁性纳米颗粒成功地将NHE1反义基因转染至SGC-7901胃癌细胞中。与亲本细胞相比,光镜下NHE1反义基因转染的SGC-7901胃癌细胞胞体增大,胞质丰富,核大小相对一致,核质比和核分裂象减少,异型性减少。透射电镜下细胞线粒体数量增多、体积增大,并可见髓鞘样变,粗面内质网、高尔基体较发达,内质网扩张,核偏向一侧,极性增强。结论NHE1反义基因磁性纳米微粒能使SGC-7901胃癌细胞发生形态学及超微结构的变化,有促进SGC-7901胃癌细胞分化的作用。Objective To study the effects of Na^+-H^+ exchanger 1 (NHE1) antisense gene magnetic nanoparticies on the morphology of SCG-7901 gastric caner cells. Methods Antisense NHE1 eukaryotic expres- sion on vector pcDNA3.1 was constructed with recombinant DNA technique and transfected into SGC-7901 gastric carcinoma cells by DCIONP. The dextran coated iron oxide nanoparticles (DCIONP) was synthesized with deposition. Then, NHE1 antisense gene magnetic nanoparticles were constructed. We examined the morphological changes of the transfected and untransfected cells under light and electron microscopes. Results DCIONP successfully transfected NHE1 antisense gene into SGC-7901 gastric cancer cells. Under the light microscope, we found that transfected SGC-7901 gastric cancer cells with NHE1 antisense gene were larger with abundant cytoplasm and comparatively same size of nuclei, and the ratio of nuclei to cytoplasm, mitotic count and heteromorphism decreased when they were compared with their parent cells. The electron microscope showed increased number of mitochondrion and size of the transfected SGC-7901 gastric cancer cells with NHE1 antisense gene, expanded Golgi apparatus, rough endoplasmic reticulum and endoplasmic reticulum and strengthened heteropolarity. Conclusion The morphology and microstructure of SGC-7901 gastric carcinoma cells change after the transfection of antisense NHE1 gene that can promote the differentiation of SGC-7901 gastric carcinoma cells.
