摘  要:目的:了解兔屈趾肌腱Ⅱ区伤口愈合过程中转化生长因子β1受体在不同时间和部位的表达情况。方法:实验于2004-09/2005-07在青岛大学医学院附属医院动物实验中心完成。①实验材料:清洁级成年新西兰大白兔42只,体质量4.0~4.5kg,雌雄不拘。②实验干预及分组:36只成年新西兰大白兔左前中趾Ⅱ区屈趾深肌腱被完全切断并修复为实验组,分别于1,7,14,21,28,56d获取肌腱,每个时间点6只;另取6只兔,不损伤和修复屈趾肌腱做对照组。③实验评估:用Western blot和免疫组织化学技术分析两组转化生长因子β1受体的表达差异。结果:①Western blot发现实验组切断修复后的肌腱转化生长因子β1受体蛋白的上调,主要集中在腱鞘、腱外膜和沿肌腱切口处,在14d达到高峰至56d才明显降低;对照组只见极少的受体表达。②免疫组织化学染色结果与Western blot是一致的。结论:肌腱损伤修复后,肌腱和腱鞘转化生长因子β1受体的表达明显增加,在术后14d达到高峰,56d开始降低,这种受体上调可能为屈指肌腱术后瘢痕形成的生物学调节提供新的途径。AIM: To analyze the temporal and spatial distribution of transforming growth factor beta (TGF-β) receptors in rabbit zone Ⅱ flexor tendon wound healing model. METHODS: The experiment was carded out in the Animal Experimental Center of Affiliated Hospital to Qingdao University Medical College from September 2004 to July 2005. ①Forty-two adult New Zealand white rabbit of either gender, aged 4.0-4.5 kg, were used for the experiment. ②Thirty-six rabbits were adopted to isolate forepaws from the middle digit flexor digitorum profundus tendon in zone Ⅱ as experiment group. The tendons underwent transection and immediate repaired, then harvested at increasing time points: 1, 7, 14, 21, 28, and 56 days postoperatively, 6 rabbits at each time point. The control flexor tendons were harvested without transection and repair in other 6 rabbits. ③Westem blot and immunohistochemical analysis were used to detect the expression for TGF-β receptors. RESULTS: ①Westem blot of the tendons in experiment group demonstrated the up-regulation of TGF-β receptor protein levels, which mainly appeared in the tendon sheath, the epitendineum and the incision along the repair site. Furthermore, the TGF-β receptor expression peaked at day 14 and decreased by day 56 postoperatively. In contrast, minimal expression was observed in the untransected and unrepaired control tendons. ② The outcome of immunohistochemical staining was similar to that of Westem blot. CONCLUSION: The tendon and epitenon have the highest TGF-β receptor expressions after injury and repair; The peak levels of TGF-β receptor expression occur at day 14 and decrease by day 56 postoperatively; The up-regulation of TGF-β receptor during flexor tendon wound healing provides new targets for biomolecular modulation of postoperative scar formation.
