摘 要:目的:探讨单基因遗传自然发病型(db/db)糖尿病小鼠颌下腺转化生长因子β1和血管内皮生长因子表达变化与糖尿病性神经病变关系。方法:实验于2003-09/2004-05在承德医学院中心实验室和承德医学院附属医院病理科完成。①实验材料:引进日本C57BL/ksj-db/+m表型正常隐性基因小鼠80只。②实验干预及分组:小鼠近亲(兄妹)交配,得到纯合子和非纯合子后代。纯合子后代为单基因遗传自然发病型糖尿病小鼠,即db/db型,相当于人类Ⅱ型糖尿病;非纯合子后代,即db/+m型,为糖尿病和肥胖基因携带者。生后4周起,平均血糖值升高达10mmol/L以上且同时伴发肥胖者确定为糖尿病鼠,选取3,4,6,8,10个月龄db/db型,血糖>10mmol/L的雄性小鼠作为实验组;相应月龄的雄性同种db/+m型小鼠为对照组,每组5只动物。③实验评估:动物麻醉多聚甲醛灌注固定后,取3,4,6,8,10个月两组小鼠颌下腺,采用SP免疫组织化学染色,观察颌下腺的组织学改变和转化生长因子β1、血管内皮生长因子阳性表达的变化。结果:除去死亡和造模失败的小鼠外,正常对照组16只、实验组25只进入结果分析。①两组小鼠内皮细胞生长因子的表达变化:内皮细胞生长因子在正常鼠及糖尿病鼠颌下腺的血管内皮细胞中均有表达,实验组与对照组内皮细胞生长因子阳性细胞表达随月龄增加而增多。在月龄相同时,实验组的表达均强于同龄对照组(P<0.05或0.01)。②两组小鼠转化生长因子β1的表达变化:转化生长因子β1在正常及糖尿病鼠颌下腺的腺泡细胞、颗粒曲管及纹状管细胞中均有表达,阳性颗粒位于胞浆内。对照组阳性细胞数随月龄变化不大,实验组阳性细胞表达数随月龄增加而增多。在月龄相同时,实验组的表达均高于同龄对照组(P<0.01)。结论:db/db糖尿病小鼠颌下腺转化生长因子β1表达增强,同时有间质纤维增强,转化生长因子β1表达�AIM:To investigate the relation of diabetic neuropathy with the expression of transforming growth factor β1 (TGF-β1) and vascular endothelial growth factor (VEGF) in submandibular gland of C57BL/ksj-db/db mutant diabetic mice. METHODS: The experiment was performed in the central laboratory and Department of Pathology, Affiliated Hospital of Chengde Medical College it from September 2003 to May 2004. ①Eighty C57BL/ksj-db/+m mice with recessive gene (Japan) were selected, and inbred to obtain the homozygote and non-homozygote offspring. Homozygote mice with monogenic inheritance diabetes, namely db/db type, were equal to having type 2 diabetes in human beings; non-homozygote mice, db/+m type, were carriers of diabetes and obesity gene. The mice with mean blood glucose 〉 10 mmol/L from the fourth week after born and complicated with obesity were regarded as diabetic ones. The 3, 4, 6, 8, 10 month-old db/db male diabetic mice served as experimental group and the age-matched db/+m mouse as control with 5 animals in each group. ② After the mice were fixed with paraformaldehyde, submandibular gland was harvested. Immunohistochemistry technology was applied to observe the histological changes of submandibular gland and positive expression of TGF-β1 and VEGF. RESULTS: Except those of died and failed to model, 16 mice in control group and 25 in experimental group were involved in the result analysis. ① Endothelial cell growth factor was expressed both in healthy mice and vascular endothelial cells of submandibular gland in diabetic mice. The expression of vascular endothelial cell was increased with the month of mice prolonged in two groups. The expression in experimental group was higher than that in control group at the same age (P 〈 0.05 or 0.01). ②TGF-β1 was expressed in the secretary striate ducts, granular tubules, and acinar cell of submandibular gland of healthy and diabetic mice.
