摘 要:目的:观察血管内皮生长因子在口腔扁平苔藓上皮细胞中的表达。方法:①实验材料:选择2005-01/2006-03在泰山医学院附属医院经临床与病理检查确诊的7例口腔扁平苔癣患者口腔黏膜损害标本,所有患者此前均未经任何治疗,无其他系统性疾病,伴有系统性疾病者不纳入研究范围。②实验方法:分离上皮层和固有层后,体外培养口腔扁平苔癣上皮细胞,提取总RNA和蛋白质,另取5例来源于本科手术切除的正常口腔黏膜作为对照,患者知情同意。③实验评估:以逆转录-聚合酶链反应检测口腔扁平苔癣和正常口腔黏膜上皮细胞血管内皮生长因子mRNA的表达,蛋白免疫印迹检测血管内皮生长因子蛋白质的表达水平。结果:①正常口腔黏膜上皮细胞和口腔扁平苔癣的上皮细胞均表达血管内皮生长因子4种亚型:血管内皮生长因子121、血管内皮生长因子145、血管内皮生长因子165和血管内皮生长因子189。其中血管内皮生长因子121、血管内皮生长因子165和血管内皮生长因子189mRNA在口腔扁平苔癣上皮细胞水平明显高于正常口腔黏膜上皮细胞的表达,差异具有显著意义(P<0.01)。②蛋白免疫印迹发现口腔扁平苔癣黏膜上皮细胞血管内皮生长因子蛋白质水平较正常口腔黏膜上皮细胞表达增加(P<0.01)。结论:血管内皮生长因子mRNA和蛋白质在口腔扁平苔癣上皮细胞中表达增高。AIM: To investigate the expression of vascular endothelial growth factor (VEGF) in the epithelial cells of oral lichen planus (OLP). METHODS: ①Samples of OLP lesions were harvested from 7 patients, who were diagnosed as OLP by clinical and pathological examination in Affiliated Hospital of Taishan Medical College from January 2005 to March 2006. All patients had not received any treatment, and they had no systemic disease. ②After the epithelial lamina and proper layer were isolated, epithelial cells from OLP lesions were cultured in vitro. The total RNA and protein were extracted. Meanwhile, the normal oral membrane excised from 5 patients surgically served as control. ③VEGF was detected at the mRNA and protein levels defined by RT-PCR and Western blot, respectively. RESULTS:①Four isoforms of VEGF, VEGF121, VEGF145, VEGF165 and VEGF189 were detected in cultured normal oral epithelial cells and OLP epithelial cells. Furthermore, the levels of VEGF121, VEGF165 and VEGF189 mRNA were significantly higher in OLP than that in normal membrane (P 〈 0.01). ②Protein levels of VEGF in OLP were remarkably higher than those in normal samples (P 〈 0.01). CONCLUSION: Expression of VEGF in OLP epithelial cells is higher than that in normal oral epithelial cells at both mRNA and protein levels.
