摘  要:目的研究雷公藤内酯醇(T10)对血管内皮细胞移行活性及组织型纤溶酶原激活物(t-PA)蛋白表达的影响,探讨T10对新生血管的抑制作用。方法体外培养传3代人脐静脉内皮细胞,加入不同质量浓度的T10,观察内皮细胞的移行活性和免疫组织化学染色检测内皮细胞t-PA蛋白表达量。结果T10可抑制内皮细胞的移行活性,使移行距离缩短;减少内皮细胞t-PA蛋白表达,与对照组相比差异有统计学意义(P<0.01)。结论T10可抑制内皮细胞移行,减少内皮细胞t-PA蛋白表达,抑制血管生成。Objective Researches have showed that neovascularization usually accompanies with inflammation and immune process, and triptolid (T10) can inhibit inflammation and immune reaction. However, effect of T10 on angiogenesis is unclear now. This study aimed to investigate the inhibitory effects of T10 on the migration of vascular endothelial cells cultured in vitro and expression of tissue-type plasminogen activator ( t-PA ) and to elucidate the anti-angiogenic mechanism of T10. Methods Human umbilical vein endothelial cells(HUVECs) from newborn were cultured in vitro and identified with Ⅶ factor staining. The passage 3 cells were treated with different concentrations of T10 ( 5,10,20,30 μg/L). Cultured cells were not treated by T10 as negative control,and dexamethasone alone was added in medium as positive control. Effect of T10 on the migration of HUVECs was assessed, and protein expression of t-PA were detected by immunohistochemical assay. Results Cultivated HUVECs showed positive reaction for Ⅶ factor. The migration distance of HUVECs was obviously shortened as the increase of concentration of T10 in comparison with control group. Expression level of t-PA protein was significantly decreased in various concentrations of T10 groups compared with negative and positive control groups ( P 〈 0.01 ). Conclusion T10 inhibits the proliferation of HUVECs and expression of t-PA,implying that it can suppress the angiogenesis effectively.
