积雪苷霜软膏对豚鼠皮肤色素沉着的预防作用

摘　　要：目的观察积雪苷霜软膏对紫外线照射攻击后豚鼠皮肤色素沉着的预防作用。方法试验豚鼠以320nm中波紫外线(UVB)照射一日1次(照射时间60分钟)共30日;试验豚鼠造模同时分别涂赋形剂(模型组)、维A酸乳膏(维A酸组)和积雪苷霜软膏(积雪苷霜软膏组)预防,观测皮肤组织酪氨酸(Tyr)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量变化,并运用Elivision^(TM) plus免疫组化法和图像分析法观测黑色寨细胞在表皮中的分布及分泌功能情况。结果各模型组豚鼠皮肤黑色素沉着增加,皮肤Tyr、MDA含量明显升高,SOD含量降低。积雪苷霜软膏组豚鼠皮肤Tyr、MDA水平显著降低,SOD水平升高;豚鼠表皮黑色素沉着面积减小、黑色素沉着程度降低。结论积雪苷霜软膏对紫外线照射攻击豚鼠所致皮肤色素沉着有一定的预防作用。Objective To investigate the prophylactic effects of asiaticoside ointment on skin the 320nm UVB radiation. Methods Guinea pigs were exposed to 320nm UVB radiation and asiaticoside ointment was topically applied on the skin simultaneously for 30 days.The skin of treated,model and control groups was collected, and the content of tyrosine（Tyr）, MDA and the activity of SOD were detected. ElivisionTM plus immunohistochemical method was used to assay the distribution and secretion of melanocytes in epidermis. Results Chronic UVB radiation induced marked increase of Tyr and MDA content and decrease of SOD activity, as well as pigmentation in guinea pig skin.The changes were inhibited by asiaticoside ointment. Conclusion Prophylactic application of asiaticoside ointment can significantly improve UVB-induced pigmentation in guinea pig＇s skin,which may be contributed to inhibition of melammocyte proliferation and melanin synthesis.
