摘 要:目的探讨胸腺修饰用于猪心脏移植免疫耐受诱导的可能性。方法将实验动物随机分为A、B、C三组,各5头。A组仅行异位心脏移植,B组在A组基础上用全身照射,C组全身照射加胸腺内注射供体脾细胞。用混合淋巴细胞反应(MLR)来了解其免疫功能的相关改变。预处理后2周,行异位心脏移植,术后检测IL-2、IL-10水平,并观察移植心脏的生存时间。结果在C组受体胸腺修饰后第2周,混合淋巴细胞反应刺激效应有所下降(P〈0.05),而A组预处理前后无显著差异,(P〉0.05)。C组供心存活期5.6 d,较A组2.4 d和B组2.2 d明显延长,(P〈0.05),而空白组与单纯照射组比较P〉0.05。实验组移植后1-3 d动物血清IL-2的水平明显低于对照组和空白组(P〈0.05),IL-10水平三组无明显差异(P〉0.05)。结论实验动物预处理后,产生了一定程度的免疫抑制,但并未获得"永久性"免疫耐受。[ Objective ] To investigate to induce immune tolerance with intrathymic inoculating allogeneic spleen cell in swines. [ Methods ] In group A (blank group), heterotopic cardiac transplant (HCT) were applied in recipient pigs without IT and WBI. In group C (IT&WBI group), synchronously, allogeneic spleen cell was injected in the thymus of the recipient pig. The mixed lymphocyte reaction was applied before intrathymic inoculation and after 1,2 weeks. Then HCT was performed in pigs of group B and C after 2 weeks of WBI. Palpation of heart beat in the abdomen was counted to learn survival time of donor heart. Recipient's serum was harvested in order to analyze IL - 2 and IL - 10. [ Results ] The results of MLR stated stimulative effect after 2 weeks of intrathymic inoculation was a notable decrease compared with the preintrathymic inoculation levels 1 week after intrathymic inoculation and that of group B. Survival time of group C ( MST 5.6 days) was significantly longer than that of group A ( MST 2. 4 days) and ( MST 2.2 days). The level of IL - 2 in group C was much lower than that of in group A and group B after transplantation 1 - 3 days. There was no differenence between group A, group B and group C in the level of IL- 10. [Conclusion] Pretreatment with IT and WBI in the outbred pig could induce "transient" immune surpression, not the "permanence" immune tolerance in the rodent.
