摘  要:目的观察Wnt信号途径关键调控分子β-catenin和糖原合成酶激酶3β(Gsk-3β)在神经干细胞(Neural stemells,NSCs)增殖分化过程的动态变化,探讨其对NSCs增殖分化的调控作用。方法由胎鼠分离培养并传代扩增NSCs,免疫细胞化学鉴定其干细胞属性及多向分化潜能,Western印迹定量检测NSCs与5%血清诱导分化122、4、487、2 h后β-catenin和Gsk-3β的动态变化。结果获得具有良好增殖生长能力与多向分化潜能的NSCs体系,β-catenin高表达于NSCs,伴随NSCs分化,β-catenin表达逐渐减弱,而Gsk-3β的表达则逐步增高。结论经典Wnt信号途径在NSCs分化过程中呈现了一个由活跃到抑制的变化,提示其与脑发育中NSCs的增殖分化密切相关。Objective To detect the dynamic changes of two key wnt signaling factor β-catenin and glycogen synthase kinase-3β(Gsk-3β) so as to investigate their roles in the process of neural stem cells(NSCs) proliferation and differentiation.Methods NSCs were cultured from fetal rat brain and identified by immunofluorescence.The expression of β-catenin and Gsk-3β were examined quantitatively by Western blotting in NSCs and at 12,24,48,72 hrs after induction by 5% fetal serum.Results A culture system of NSCs with vigorous proliferation and multipotential differentiation ability was established.In the process of NSCs differentiation,the expression of β-catenin decreased with increasing expression of Gsk-3β.Conclusions The classical wnt signal pathway shows a dynamic change from activation to inhibition.It probably plays a role in the proliferation and differentiation process of NSCs.
