兔胰岛细胞团颅内移植治疗糖尿病的研究

摘　　要：将新鲜制备的成年免胰岛详细胞团（ICCs）经腰椎穿刺注入脊髓腔，采取小剂量（胰岛数为2×103～10×103）多次颅内移植治疗糖尿病兔。术后不用免疫抑制剂，不用外源性胰岛素。结果10只糖尿病免在胰岛细胞团颅内移植后，3只显效，2只有效，无排斥反应及神经系统异常。组织学检查显示，受体免原胰腺萎缩，脑脊髓组织形态正常，移植胰岛分别种植在大脑、脑室及脊髓表面，部分融合成难生长，周围无免疫细胞及外分泌腺泡组织。本实验为糖尿病的治疗提供了一个新思路。Multi-intrathecal allo-transplantation of islet-like cell cluster (ICCs, islets: 2×103~ 10×103) from multi-donors into alloxan-diabetic rabbits significantly reduced the blood sugar level in 50% recipients (P＜0. 05). 30% of them remained in the state of normoglycemia for up to 2 years. No rejection response and abnormalities were found in CNS of patients. Histological examination showed presence of a lot of islet clusters on the surfaces of brain, ventricles and spinal cord but absence of lymphocytic infiltration. Our study provided a new approach for the treatment of diabetes.
