摘  要:目的观察艾灸调节肿瘤免疫抑制效应与IL-2受体-Jak-Stat信号转导通路的关系,探讨艾灸调节免疫抑制效应的作用机制。方法将50只Balb/c小鼠随机分为正常对照组、正常艾灸组、荷瘤对照组、艾灸治疗组和艾灸非经穴组等5组,每组各10只。观察艾灸大椎(2mg/壮,2壮/次,共6次)对H22移植性实体瘤诱导免疫抑制效应的影响。采用125I-IL-2放射配基方法观察脾淋巴细胞高亲和IL-2受体数量及亲和力,RT-PCR方法观察脾细胞IL-2受体各亚基及信号分子Jak1、Jak3、Stat5a、Stat5b mRNA表达水平。结果荷瘤对照组脾淋巴细胞中亲和力IL-2R数量明显低于正常对照组;艾灸治疗组中亲和力IL-2R数量较荷瘤对照组明显增加,荷瘤对照组脾淋巴细胞IL-2Rα、IL-2Rβ、Jak1、Stat5a、Stat5b mRNA表达水平与正常对照组比较明显降低;艾灸治疗组IL-2Rα、IL-2Rβ、Jak1、Stat5a、Stat5b mRNA表达水平明显高于荷瘤对照组。各组IL-2Rγ mRNA、Jak3 mRNA表达水平未见显著差异。结论针灸正向调节肿瘤机体非特异性免疫功能低下或受抑状态,与上调淋巴细胞IL-2受体数目及其IL-2Rα、IL-2Rβ和相关信号分子Jak1、Stat5a、Stat5b表达水平有关;但研究结果并未显示出针灸调节肿瘤诱导的免疫抑制效应与Jak3和IL-2Rγ链相关。Objective To investigate the molecular mechanism of moxibustion in modulating immunosupression by observing IL-2 receptor-Jak-Stat signal transduction in tumor-bearing mice. Method Fifty Balb/c mice were randomly divided into 5 groups: normal-control group, treatment-control group, tumor-control group, treatment group and non-acupoint treatment group. H22 tumor-bearing mice were treated by moxa on Dazhui(GV 14),2mg/cone,2 cones per day, 6 times in total. The amount of high affinity IL-2 receptor in splenic lymphocytes was determined by 125^I-IL-2 Radio Binding Assay (RBA). The expression of IL-2R subunit mRNA and Jakl, Jak3, Stat5a, Stat5b mRNA were studied by RT-PCR. Result There was a significant decrease of mediate affinity IL-2 receptor in the tumor-control group than that in the normal-control group. The amount of mediate affinity IL-2 receptor in the treatment group was increased remarkably, when compared with that in the tumor-control group. The expression level of IL-2Rα, IL-2Rβ mRNA, Jak1 mRNA, Stat5a and Stat5b mRNA of splenic lymphocyte in the tumor group were lower than that in the normal group. The expression level of IL-2Rα, IL-2Rβ mRNA, Jak1 mRNA, StatSa, 5b mRNA was significantly increased after moxibustion treatment. There was no statistic difference in the expression level of Jak3 and IL-2Rγ mRNA among all five groups. Conclusion Moxibustion can modulate immunosuppressive state induced by tumor related to the up-regulation of the amount of mediate affinity IL-2 receptor and Jak-Stat signaling.
