摘 要:目的探讨对氧磷酶(PON)中PON155M/L和PON2148A/G基因多态性与高血压的关系。方法采用PCR-RFLP方法检测201例高血压组和94例对照组的PON155M/L和PON2148A/G基因多态性。结果与对照组比较,高血压组血浆PON、总超氧化物歧化酶(T-SOD)活性明显降低,丙二醛(MDA)浓度显著升高;PON155LM基因型、M等位基因频率,PON2148GG/AG基因型、G等位基因频率较对照组明显增多;PON155LM基因型的PON和T-SOD活性较LL基因型明显降低,而收缩血压、舒张血压显著增高;PON2148AG和GG基因型的PON活性较AA基因型者明显降低,GG基因型的MDA、AG基因型的收缩压和舒张压则显著升高。logistic回归分析显示PON155LM基因型、PON2148GG/AG基因型是高血压的危险因素。结论PON155M等位基因、PON2148G等位基因是高血压的危险因素,此等位基因携带者血浆PON活性降低。Objective To study the associations of PON, 55 M/L and PON2 148 A/G genetic polymorphisms with essential hypertension(ET). Methods PCR-RFLP method was used to identify genotypes of PON1 55 M/L and PON2 148 A/G genetic polymorphisms in 201 ET patients and 94 controls. Results Compared with controls, the plasma activities of PON and T-SOD decreased obviously, while plasma MDA concentration increased markedly in hypertensives. There were more LM genotype and M allele of PON, 55 M/L, GG and AG genotypes and G allele of PON2 148 A/G in hypertensives than in controls. The activities of plasma PON and T-SOD were lower, but SBP and DBP were higher in individuals with PON, 55 LM genotype than in those with LL genotype. The activity of plasma PON was reduced, while plasma MDA concentration, SBP and DBP increased in individuals with PON2 148 GG/AG genotypes than in those with AA genotype. Logistic regression analysis demonstrated that PON, 55 LM genotype and PON2 148 GG/AG genotype were independent risk factors for ET. Conclusion PON1 55 M allele and PON2 148 G allele are risk factors for essential hypertension, furthermore, the plasma activity of PON also reduces obviously in individuals with these alleles.
