蜕膜巨噬细胞TSP-1 IL-10及IFN-γ的表达及其相互关系探讨

摘　　要：目的探讨蜕膜巨噬细胞凝血酶敏感蛋白-1(TSP-1)、白介素-10(IL-10)及干扰素-γ(IFN-γ)的表达。方法对2005年7月至2006年7月在上海交通大学附属仁济医院计划生育门诊要求人工流产的正常妊娠妇女20例(A组)和10例原因不明复发性流产(URSA)患者(B组)检测蜕膜TSP-1mRNA、蜕膜巨噬细胞表面TSP-1的表达及蜕膜巨噬细胞分泌IL-10和IFN-γ的量,并在培养液中加入相应刺激物,观察细胞因子分泌量的变化。结果与A组相比,B组蜕膜TSP-1mRNA表达明显降低(P<0·05)。蜕膜巨噬细胞TSP-1表达水平明显降低(P<0·01)。蜕膜巨噬细胞分泌IL-10的量显著降低(P<0·05);在培养液中加入各种干预因素对两组病例蜕膜巨噬细胞分泌IFN-γ无显著影响;加入TSP-1后,两组蜕膜巨噬细胞分泌IL-10的量均增加(P<0·05);加入抗TSP-1后A组蜕膜巨噬细胞分泌IL-10的量降低(P<0·05)。结论URSA患者蜕膜TSP-1表达下降,且蜕膜巨噬细胞分泌IL-10的量降低,可能是导致URSA发病的原因。TSP-1可能在调控蜕膜巨噬细胞分泌IL-10的过程中起重要作用。Objective To investigate the expression of TSP-1 on decidual macrophages and its effect on the secretory function of IL-10, IFN-γ on macrophages. Methods There were 20 women with selective termination of normal early pregnancy（ group A）and 10 women with unexplained recurrent spontaneous abortion （group B）that were involved in the research. Detect the expression of TSP-1 mRNA in deciduas and the expression of TSP1 on decidual macrophages and cytokine secretion of decidual macrophages. Some stimulators were respectively added into the culture fluid, to observe the changes of secretory volume of decidual macrophages. Results Compared with group A, the expression of TSP-1 mRNA on decidual macrophages of group B was significantly decreased （ P 〈 0. 05 ）. The expression of TSP1 was decreased significantly （P 〈 0. 01 ） on decidual macrophages. The secretory volume of IL-10 in decidual macrophages was significantly higher（ P 〈 0. 05 ）. The secretory volume of IL-10 in decidual macrophages of group 1 and group 2 were elevated when TSP-1 was added in culture fluid（P 〈0. 05）. The secretory volume of IL-10 in decidual macrophages of group 1 was decreased when anti-TSP-1 antibody was added in culture fluid （ P 〈 0. 05 ）. Conclusion The expression of TSP-1 on decidual macrophages of women with URSA is decreased, and the secretory volume of IL-10 in decidual macrophages is also decreased. This may be the reason of URSA. TSP-1 may play an important role in regulating the secretory function of IL- l0 in decidual macrophages.
