摘　　要：将牛ＦＳＨ－βｃＤＮＡ连接于ｐＭＭＴ中金属硫蛋白（ＭＴ）启动子下游，与带有二氢叶酸还原酶（ＤＨＦＲ）基因的质粒共转染中国仓鼠卵巢细胞ＣＨＯ－ｄｈｆｒ－，在ＤＨＦＲ＋转化细胞中所整合的ＭＴ－ｂＦＳＨ－β基因可被重金属镉诱导。经１０μｍｏｌ·Ｌ－１ＭＴＸ加压后，获得可表达、分泌ｂＦＳＨ－β的细胞株，其中一株ｂＦＳＨ－β的产率２４ｈ可达７００ｎｇ·Ｌ－１×１０６ｃｅｌｓ，并能稳定分泌。表达产物经ＳＤＳ－ＰＡＧＥ，Ｗｅｓｔｅｒｎ－ｂｌｏｔ分析表明：所表达ｂＦＳＨ－β的分子量同天然产品相同，为１４０００Ｄ，并具有免疫活性。bFSH β cDNA has been cloned in the mammalian cell expression vector pMMT, under the control of mouse metallothionein I(MT) promoter,and cotransfected the CHO dhfr - cells with pSV 2 dhfr. The MT bFSH β gene intergrated with dhfr on the chromosomes of transformed cells could be induced by heavy metal(CdCl 2), and also be coamplified by methortrexate(MTX). When treated with 10 μmol·L -1 MTX, stable cell lines were obtained that could secret bFSH β up to 700 ng·L -1 ×10 6 cells/24 h. SDS PAGE and Western blot showed that the Molecular Weight of the excreted recombinant bFSH β was the same as that of natural product which is 14 000 daltons.
