摘 要:目的观察翻白草水提液(HPD)对高脂膳食加链尿佐菌素(STZ)诱导的2型糖尿病(2-DM)胰岛素抵抗大鼠血清胰岛素水平及白色脂肪组织抵抗素基因mRNA表达的影响,探讨HPD提高胰岛素敏感性的可能机制。方法选用高脂饲料诱导的Wistar大鼠建立胰岛素抵抗模型后,继续用小剂量STZ建立2-DM胰岛素抵抗模型。将造模大鼠随机分为模型对照组(水)和3个HPD组(3g/kg,6g/kg,12g/kg),另选10只大鼠为正常对照组。各组灌胃8周后,空腹取材,放射免疫法测定空腹血清胰岛素,RT-PCR法检测肾周脂肪组织抵抗素(resistin)基因mRNA水平。结果与模型对照组比较,HPD高、中剂量组能显著降低空腹血清胰岛素水平;HPD各组均能显著降低大鼠肾周脂肪组织抵抗素基因mR-NA表达。结论HPD水提液具有改善2-DM胰岛素抵抗大鼠的胰岛素抵抗作用。Objective: To observe the effects of Herba Potenillae Discoloris decoction on resistance gene mRNA expression of serum insulin and white adipose tissue of type 2 diabetes insulin resistance rats induced by high - fat diet and streptozotocin, to study possible mechanism of HPD enhancing insulin sentivity. Methods: To build model of type 2 diabetes insulin resistance rats induced by high - fat diet and streptozotocin. The model rats were divided into four groups at random : model group and low, middle, high dosage of HPD groups with 10 in each group, another 10 rats were selected as normal group. Serum insulin was detected by radioimmunossay and mRNA expression of insulin resistance Gene was determined by RT - PCR after rats were fed eight weeks. Results: Compared with the model group, middle, high dosage of HPD discolofis can reduce the level of serum insulin obviously ; low, middle, high dosage of HPD can reduce resistance gene mRNA expression of adipose tissue obviously. Conclusion : Herba Potenillae Discolofis decoction can improve insulin resistance of type 2 diabetes insulin resistance rats.
