摘  要:[目的]为了探索一种快速、简便,能够用于SSR分析的玉米DNA提取法。[方法]采用改进的常规DNA提取法和DNA快速提取法提取玉米种子胚和黄化苗叶片中的DNA,进行SSR分析,通过琼脂糖电泳和聚丙烯酰胺凝胶电泳对SSR产物进行检测。[结果]用常规法提取时,用氯仿∶异戊醇(24∶1)抽提1次就可获得质量好和浓度高的DNA;无须抽提,直接吸取上清液,可得到高浓度的DNA,也能满足SSR分析的需要;同样的提取法,SDS的提取效果比CTAB好。对快速提取法进行分析表明,相同的提取液和提取方法,采用种子胚提取的DNA浓度和质量明显高于叶片DNA。[结论]该研究为SSR在玉米种质及纯度鉴定上的应用提供了技术上的保证,有利于该项技术的普及和推广。[Objective] The research aimed to investigate a rapid and simple extraction method of maize DNA, which could be used in SSR analysis. [Method] Using improved conventional DNA extraction method and rapid DNA extraction method, DNA in maize seed embryo and the yellow seedlings were extracted to analyze by SSR and its amplification products were detected by agarose electrophoresis and polyacrylamide gel electrophoresis. [Result] Extracted by conventional method, DNA with good quality and high concentration could be obtained from one-time chloroform/ isoamyl alcohol extraction. DNA with high concentration could be directly obtained from supernatant fluid without extraction, which also could meet the need of SSR analysis. By the same extraction method, the extraction effect with SDS was better than that with CTAB. The analysis on rapid extraction method indicated that with the same extracted liquid and the same extraction method, the DNA concentration and quality extracted from seed embryo were higher than those from leaves DNA. [Conclusion] The research provided the technical guarantee for the SSR application in identifying the maize germplasm and purity, which was favorable for the popularization and extension of SSR teehnology.
