摘  要:目的:获取分泌结合力和特异性均好的抗人促甲状腺素(hTSH)单克隆抗体的杂交瘤细胞株.方法:用人垂体提取液为免疫原,免疫BALB/c小鼠,以聚乙二醇为融合剂,亚克隆三次.结果:获得2株分泌与hTSH特异性结合的单克隆抗体杂交瘤细胞株,所分泌的免疫球蛋白为IgG1,液相中与hTSH的最大结合率分别为60%和45.1%.经位点分析,2株单克隆抗体分属不同位点.结论:①人垂体提取液可用于制备hTSH单克隆抗体;②此2株单克隆抗体的获得为建立敏感的hTSH测定方法奠定了基础.Objective: To get the hybridomas cell lines secreting anti thyrotropin monoclonal antibodies with high affinity and specificity. Method: BALB/c mice was immunized with extract of human pituitary glands. The spleen cells of the immunized mouse were fused with mouse myeloma cells in polyethylene glycol and the positive clones were subcloned 3 times. Results: Two hybridomas cell lines which secrete anti thyrotropin monoclonal antibodies with high affinity and specificity have been collected. The antibodies were of the IgG 1 subclass and their maximum binding with thyrotropin were 60% and 45.1% respectively. By competitive binding studies, the pair monoclonal antibodies directed against different epitopes of human thyrotropin. Conclusion: (1) The extract of human pituitary glands could be used to produce monoclonal anti pituitary hormones antibodies; (2) The two anti thyrotropin monoclonal antibodies produced in this study could be used in the establishment of sensitive measurement of human thyrotropin.
