摘  要:目的探讨VEGF基因治疗和MSCs移植各对梗死心肌组织的影响。方法将相同基因背景的SD雄性大鼠30只随机分成A、B、C三组。建立心肌梗死模型后,MSCs组注射MSCs 4×106cells/50μl。AdVEGF组注射AdVEGF 5×107pfu 50μl。Control组(对照组)注射无血清IMDM培养液50μl。移植前、移植后2、4周分别用超声检测心功能。病理中检测细胞存活分化和血管新生情况。结果MSCs组移植2、4周后AdVEGF组与Control组相比,心功能较移植前有明显改善(P<0.01),而且病理提示大部分MSCs己向心肌样细胞分化。MSCs组和AdVEGF组都有促血管新生作用,MSCs组为(5.94±1.67)mm;AdVEGF组(13.59±1.08)mm,Control组(对照组)无新生血管出现。结论移植于梗死心肌的骨髓间充质干细胞可以存活,并在一定程度上改善了受体的心功能。Objective To investigate effect of vascular endothelial growth factor (VEGF) gene therapy and mesenchymal stem cells (MSCs) transplantation on infracted myocardium in rats. Methods Thirty SD rats were randomized into three groups. In group A,4× 10^6 cells/ 50 μl of MSCs were implanted into infracted myocardium (n = 10). In group B,5×10^7 pfu 50 μl of Ad VEGF were injected into infracted myocardium (n = 10). In group C,only 50 μl serum free medium were injected into infracted myocardlum ( n = 10). Before transplantation,2 weeks and 4. weeks after transplantation,the cardiac function was followed up by echocardiography, and cell identification and angiogenesis evaluation were detected. Results Compared with control group, the cardiac functions in group A improved significantly 2 weeks and 4 weeks after transplantation ( P 〈 0.05 ). Histologic results showed that most of the MSCs were immature like fetal cardiomyocytes. Angiogenesis was occurred in group A and group B. (5.94±1.67,13.59 ± 1.08 for group A, B, respectively ). No angiogenesis was presented in control group. Conclusion MSCs which implanted into infracted myocardium can survive,and enhance the cardiac function in infracted myocardium tissue.
