摘 要:目的研究丹参水提液中与脂质体模拟生物膜的相互作用。方法应用平衡透析与高效液相色谱-质谱联用技术对丹参水提液中与脂质体模拟生物膜有相互成分进行分析、鉴定。同时考察了透析时间、脂质体浓度和pH值对丹参与脂质体相互作用的影响。结果丹参水提液中有8个成分与模拟生物膜相互作用明显,经在线HPLC-MS分析、鉴定,其中5个成分分别为原儿茶酸、咖啡酸、迷迭香酸、丹酚酸B和丹酚酸A。脂质体浓度对丹参与脂质体相互作用影响较大,随浓度增加,可与脂质体结合成分的色谱峰面积减少量明显增加,符合被动吸收规律;pH值对其中5个成分的吸收有明显影响。结论采用脂质体平衡透析和高效液相色谱-质谱联用的方法,能方便、有效地预测丹参在体内的吸收情况,为丹参的药效物质基础研究提供依据。 OBJECTIVE To study the interaction between water-soluble constituents in Radix Salviae Miltiorrhizae and biomembrane model. METHODS Using balanced dialysis followed by HPLC/MS analysis, the water-soluble constituents in Radix Salviae Miltiorrhizae which interacted with liposome were analyzed and identified. The experiment conditions were investigated, including the concentration of liposome, the pH value of buffer, and balance time. RESULTS The areas of eight water-soluble constituents of Radix Salviae Miltiorrhizae were decreased obviously. Among them, five peaks were identified as protocatechuic aldehyde,caffeic acid, rosmarinic acid,salvianolic acid B and salvianolic acid A by LC-ESI/TOF MS. The concentration of liposome and the pH value of buffer remarkably affected the absorption of most constituents. CONCLUSION The developed method can simply and effectively estimate the absorption of Radix Salviae Miltiorrhizae in vivo.
