摘 要:目的:分析比较炮制与提取方法对黄芩药材中活性成分提取的影响。方法:以黄芩生药材和炮制药材为分析对象,比较5种不同提取方法对黄芩苷、汉黄芩苷、黄芩素、汉黄芩素提取的影响。色谱条件Agilent Zorb-ax Extend-C18色谱柱(4.6 mm×250 mm,5μm);流动相甲醇-乙腈-水-乙酸(18∶25∶57∶0.2);流速0.8 mL.min-1;检测波长275 nm;柱温30℃。结果:生药材酶法提取黄芩素、汉黄芩素得率最高,分别为5.41%,1.30%;60%乙醇回流提取黄芩苷、汉黄芩苷得率最高,分别为10.11%,3.55%。炮制药材不同提取方法的效果接近,以酶法提取效果最好,黄芩苷、汉黄芩苷和黄芩素、汉黄芩素得率分别为10.63%,3.60%和1.81%,0.59%。结论:对于黄芩生药材,不同提取方法对4种活性成分的提取影响较大;对于黄芩炮制药材,不同提取方法对4种活性成分的提取影响较小,针对不同活性成分应选择最适提取方法。Objective: To compare and analyze the effects of the contents of the active components in Radix Scutellariae by different processing and extracting methods. Method: The raw and processed Radix Scutellariae were used, and the concentration of baicalin, wogonoside, baicalein, wogonin were determined by HPLC after five extracting techniques. HPLC method was performed on methanol-acetonitrile-water-acetic acid ( 18: 25: 57:0. 2) as the mobile phase at 30℃ ; The chromatographic column was Agilent Zorbax Extend-C18 (4. 6 mm× 250 mm, 5 μm) ; The flow rate was 0. 8 mL · min^-1 , and the detection wavelength was 275 nm. Result: the ratios of baicalein and wogonin in raw material were 5.41%, 1.30% respectively by enzymatic extraction, which is higher than other extracting methods by raw material; The ratios of baicalin and wogonoside in raw material were 10. 11% and 3. 55% by ethanol of 60%, which is higher than other extracting methods ; Different extracting methods have no evident effects on processed materials, enzymatic extraction is the best. The ratio of baicalin, wogonoside and baicalein, wogonin is 10. 63%, 3.60% and 1.54%,0. 59%. Conclusion: Different methods have evident extracting effects on the four active components with raw material, but have no evident effects with processed material. According to different active components most suitable extracting method should be adopted.
