限制性制动方法建立心理应激失眠大鼠模型的可行性研究

摘　　要：目的探讨限制性制动方法建立大鼠心理应激性失眠模型的可行性。方法采取限制性制动方法、行为学测量、多导睡眠描记、放射免疫法测量和高效液相色谱分析。结果与对照组比较，限制型制动模型大鼠造模后觉醒时间增加、非快眼动（NREM）睡眠时间和8波减少；造模后第15天模型大鼠NREM睡眠8波比例减少[模型组：（49.08±11.33）％；对照组：（53．30±8．76）％]，睡眠觉醒次数转换增加[模型组：（41．00±16．84）次；对照组：（33．63±8．42）次]；体质量下降14．07％（P〈0．05）、旷场实验中水平活动减少60．49％（P〈0．01），垂直活动减少71．91％（P〈0．01），鼠尾悬挂实验中悬尾后的不动时间增加49．46％（P〈0．01），血清ACTH、CORT、B-EP、E和NE分别增加87．02％（P〈0．01）、46．05％（P〈0．05）、39．89％（P〈0．01）、470．54％（P〈0．01）和17．78％（P〉0．05）。结论限制性制动是建立大鼠心理应激性失眠模型可行性的方法。Objective To investigate the feasibility of developing psychological stress insomnia model of rats by restricting activity. Methods Restriction of activity, behavioral test, polysomnogram recording, radioim-munity, and high performance liquid chromatogram analysis were used in this study. Results Compared with control, the ratio of non-rapid eye movement （NREM） sleep and its delta wave decreased after restriction ; On day 15, the ratio of delta wave in NREM sleep decreased （49.08±11.33 in model group vs 53.30±8.76 in con-trol） ; the number of wake increased （41.00±16.84 in model group vs 33.63±8.42 in control）; the body weight of model group decreased by 14.07 percent （ P〈 0.05 ） ; ambulation reduced by 60.49 percent （ P〈 0.01 ） and the rearing reduced by 71.91 percent（ P 〈 0.01 ）in open-field test. In rat suspend tail test, the immobility time increased by 49.46 percent （ P〈 0.01 ）. Moreover, the serum ACTH, CORT, [β-EP, E and NE increased by 87.02 percent （ P 〈 0.01 ） ,46.05 percent （ P 〈 0.05 ） ,39.89 percent （ P 〈 0.01 ） ,470.54 percent （ P 〈 0.01 ） and 17.78 percent （ P 〉 0.05 ） respectively on day 15. Conclusion Restriction of activity is a feasible way of developing psychological stress insomnia model of rats.
