摘  要:流感病毒是威胁人类健康的重要传染性病原之一,20世纪已发生3次流感世界大流行。人们一直期望能有高效广谱的流感疫苗,这也成为了科学家研究的重要课题。1998年由安德鲁.法尔和克雷格.梅洛发现的RNA干扰(RNAi)是重要的基因沉默机制,它对病毒RNA也有很强的降解作用。RNAi也给流感病毒感染的基因治疗研究带来了新的曙光,如今科学家们在体外体内实验中都证明了RNAi可以高效抑制多种亚型流感病毒复制。RNAi不仅具有高特异性、高效率、长效性,而且应用对象广泛、成本低、使用简便,在不久的将来它将有望成为一种很有应用前景的抗流感武器。Influenza virus is one of important infectious pathogen threatening human health, worldwide influenza pandemic has already occurred three times since last century. People is expecting influenza vaccine with broadspectrum and high efficacy,which is becoming the important topic of researchers. In 1998, Andrew Fire and Craig Mello discovered that RNA interference is the important gene silencing mechanism, which has strong degradation on vires RNA and bring new idea for gene therapy of influenza vires. Experiments in vitro and in vivo ,showed that RNAi can inhibit duplication of its many subtypes. RNAi will become a useful anti-vires weapon with wide prapectire because of its high specificity, high efficacy, prolonged action, low cost and convenient application besides its wide apphcation object.
