摘 要:目的探讨适于研究脑实质大分子物质引流的大鼠尾壳核大分子示踪剂的注法。方法将SD大鼠随机分为剪断针头组、非剪断针头4μl组和非剪断针头1μl组,在立体定位仪控制下用微量进样器向尾壳核内注射伊文思蓝白蛋白复合物溶液,于72h留取脑标本并行冰冻切片,分别进行大体观察和荧光显微镜观察。结果剪断针头法伊文思蓝白蛋白复合物的注射量不准确且不稳定,而非剪断针头法注射的量准确而稳定。结论非剪断针头尾壳核示踪剂注射法经济、准确、稳定,适于脑实质内大分子物质引流的研究。Objective To discuss injection methods that macromolecular tracer entered rat caudate-putamen(CPu) in order to find a better way to study the drainage pathway from the parenchyma of the brain. Methods Adult Sprague-Dawley rats were randomly divided into cutting needle group, non-cutting needle 4ul group and non-cutting needle lul group. Evans Blue-labeled albumin(EBA) solution was injected into the rat CPu with microsyringe under stereotaxic apparatus' control. Frozen sections of rat brain samples were made and observed by naked eye and fluorescence microscope respectively. Results The cutting needle way by which tracer' s dose is injected into CPu was inaccurate and unstable, however, non-cutting needle way was accurate and stable. Conclusion Non cutting needle wethod by which tracers were injected into rat CPu was economical, accurate, stable and suitable for researching drainage of macromolecules from the parenchyma of the brain.
