摘 要:目的:探讨他莫昔芬对肾组织纤维化的影响及作用机制。方法:雌性SD大鼠,建立单侧输尿管梗阻模型(UUO),设置3组:①模型组(n=6),行左侧输尿管结扎术,0.3m l无菌等渗盐水灌胃,每天2次;②治疗组(n=6),他莫昔芬5mg/kg(0.3m l无菌等渗盐水配置)灌胃,每天2次;③假手术组(n=6)。术后第9 d处死各组大鼠。采用光镜和免疫组化观察肾间质纤维化、炎细胞浸润和α平滑肌肌动蛋白(α-SMA)表达的变化。结果:①治疗组大鼠肾间质纤维化程度较模型组明显减轻。②模型组第9 d时大鼠肾组织α-SMA表达明显增强,采用他莫昔芬治疗后,肾组织α-SMA表达的异常增强能得到有效抑制,肾组织炎细胞浸润明显减少。结论:昔芬类药物具有减轻肾间质纤维化的作用,而这一作用与其能有效阻抑肾组织上皮细胞-间充质细胞转分化和减少肾间质炎细胞浸润有关。Objective :To investigate the effects of Tamoxifen on renal interstitial fibrosis in rat. Methods :Female SD rats were divided into 3 groups randomly: (1)UUO group(6 rats), (2)treatment group (6 rats) : UUO + Tamoxifen per kg lOmg/day by gastrogavage, (3)sham operation group(6 rats). Kidneys were harvested at day 9 after UUO. The renal tissues were examined by HE, PAS and Masson. Tissue α-SMA protein expression was determined by Immunohistochemistry. Results:UUO at 9 days significantly activated α-SMA protein expression, induced interstitial fibrosis and tubulointerstitial lesion in obstructed kidney of UUO group compared with sham operation group. Conversely daily administration of Tamoxifen significantly inhibited α-SMA expression and decreased interstitial fibrosis and tubulointerstitial lesion in obstructed kidney of treatment group. Conclusion:Tamoxifen had beneficial effects on interstitial fibrosis in SD rats with UUO, and this effect may be associated with Tamoxifen inhibits α-SMA expression and inflammatory cell infiltration.
