听源性惊厥易感大鼠惊厥和点燃后前脑结构内的c-fos表达  被引量:3

C FOS EXPRESSION IN THE FOREBRAIN STRUCTURES AFTER SINGLE AND KINDLED SEIZURES IN AUDIOGENIC SEIZURES PRONE RATS

摘  要:为探讨听源性惊厥点燃和前脑结构的关系,用免疫细胞化学方法结合体视学分析,研究了Wistar种系的听源性惊厥易感大鼠(P77PMC)惊厥和点燃后,前脑结构内c-fos表达的差异。结果显示,(1)正常Wistar大鼠接受一次强音刺激后,海马、齿状回、杏仁核、内嗅皮质、嗅周皮质和额-顶皮质内未见Fos阳性神经元;(2)P77PMC大鼠一次惊厥后,除海马、齿状回外,上述被检各区内可见广泛的Fos阳性神经元,其分布具有区域差异;(3)P77PMC大鼠点燃后,海马、齿状回内有大量Fos阳性神经元,其他的被检各区内,Fos阳性神经元数量更多,阳性神经元的光密度更大,与一次惊厥后相比较,具有显著差异(P<0.01)。在点燃大鼠海马结构中,齿状回内的Fos免疫反应最强,海马CA1、CA2区次之,CA3区较弱。结果表明,听源性惊厥点燃可以导致海马结构参与到惊厥活动之中。In order to explore the relationship of audiogenic kindling and the forebrain structures, the present study was designed to investigate the difference of c fos expression in the forebrain structures after single (AS) and kindled (KAS) audiogenic seizures in P77PMC rats, a strain of Wistar audiogenic seizures prone rats. Using immunocytochemistry of Fos protein combined with morphometry, we obtained the following results:(1) Fos positive neurons were hardly detectable in the hippocampus, dentate gyrus, amygdala, entorhinal and the fronto parietal cortex in normal Wistar rats exposed to acoustic stimules one hour later; (2) in AS rats, a single audiogenic seizure induced c fos expression in all examined regions except the hippocampus and dentate gyrus, and their distribution showed regional difference; (3) in KAS rats, c fos expression was strongly induced in the hippocampus and dentate gyrus. In other examined regions of the forebrain structures, the density and intensity of positive neurons were significantly increased as compared with AS group ( P <0 01). In the hippocampal formation, dentate gyrus was the most intensely reactive, followed by CA1 and CA2 areas, whereas staining in CA3 area was low. These results show that audiogenic kindling induced by repeated acoustic stimulus recruits the hippocampal formation into the seizure network.
