摘  要:目的:同时测定注射用丹红(粉针剂)中丹参素、原儿茶醛、羟基红花黄色素 A 和丹酚酸 B 的含量。方法:采用高效液相色谱法,色谱柱:Luna C_(18)柱(250 mm×4.6 mm,5μm);流动相:0.05%磷酸水溶液(A)-乙腈(B),梯度洗脱程序为:0~80min,A:100%→67%,B:0%→33%;流速:1.0 mL·min^(-1);检测波长:280 nm。结果:建立了同时测定注射用丹红(粉针剂)中4种成分含量的方法。丹参素、原儿茶醛、羟基红花黄色素 A 和丹酚酸 B 的线性范围分别为56.3~338μg·mL^(-1)(r=0.9997),7.92~47.2μg·mL^(-1)(r=0.9996),16.4~98.2μg·mL^(-1)(r=0.9997),32.0~193μg·mL^(-1)(r=0.9999);平均回收率(n=9)分别为99.7%,99.3%,99.2%,98.8%。结论:该方法简便、准确,重复性好,为注射用丹红(粉针剂)的质量控制提供了定量方法。Objective: To determine protocatechuic aldehyde, hydroxysaffl 4 bioactive components in Danhong powder in injection, namely danshensu, ower yellow A and salvianolic acid B quantitively. Methods:The RP - HPLC method was applied and the separation was performed on a Luna C18 analytical column (250 mm × 4. 6 mm,5μm) with the mobile phase consisting of 0. 05% phosphoric acid and acetonitrile with gradient elution mode at the flow rate of 1.0 mL·min^-1. The gradient condition was 0 -80 min,0.05% phosphoric acid 100%→67% ,acetonitrile 0%→33%. The detection was set at 280 nm. Results:This method was applied to determine 4 compounds in Danhong powder for injection successfully. The standard curves were linear over the ranges of 56. 3 -338 μg ·mL^-1 for danshensu (r =0. 9997) ;7.92 - 47.2 μg ·mL^-1 for protocatechuic aldehyde( r = 0. 9996) ; 16. 4 - 98. 2 μg ·mL^-1 for hydroxysafflower The average recoveries (n acid B were 99. 7% , 99.3 with a good reproducibility yellow A(r =0. 9997)and 32.0 -193μg ·mL^-1 for salvianolic acid B(r =0. 9999). =9) %,9 and of danshensu, protocatechuic aldehyde, hydroxysafflower yellow A and salvianolic 9. 2% and 98. 8%, respectively. Conclusion: The method is simple and accurate can be used as a quality control method for Danhong powder for injection.
