摘  要:为系统研究运动心脏降钙素基因相关肽(CGRP)的变化特征,将雄性SD大鼠分为耐力训练组(E组)和对照组(C组)。E组经10周跑台训练建立运动心脏动物模型。建模成功48h后,各组抽取半数动物进行力竭运动。用酶联免疫法检测血CGRP含量,放射免疫法检测心室肌CGRP含量,免疫组织化学法和实时荧光定量PCR分别检测背根神经节CGRP免疫反应和CGRP mRNA表达。结果表明,E组血清CGRP、心室肌CGRP含量和背根神经节CGRP免疫反应显著增加,但背根神经节CGRP mRNA表达显著降低;力竭运动后,E组与C组相比,血清CGRP显著提高,心室肌CGRP含量减少,背根神经节CGRP mRNA表达轻度上调。结果提示,运动心脏重塑后CGRP合成、储备和分泌增强,但基因表达下调。力竭运动时运动心脏CGRP基因表达稳定,释放能力强。The purpose of this study was to comprehensively investigate the characteristics of calcitonin gene-related peptide (CGRP) in exercise-induced cardiac remodeling. Male SD rats were divided into two groups:endurance training group (EG) and sedentary control group (CG). 48 h after the animal model establishment by graded treadmill training lasting 10 weeks, half rats of each group were subjected to exhaustive treadmill running. The levels of CGRP in serum were determined by enzyme immunoassay, concentration of CGRP in left ventricular myocardium by radio immunoassay, CGRP-immunoreactivity in dorsal root ganglion by immunohistochemistry, the expression of CGRP mRNA in dorsal root ganglion by real-time fluorescence quantitative PCR. The result showed that in EG, content of CGRP in serum, left ventricular myocardium and dorsal root ganglia increased significantly, but gene expression in dorsal root ganglia decreased significantly. After exhaustive exercises, serum CGRP significantly increased and content in left ventricular myocardium decreased and gene expression in dorsal root ganglia was lightly up-regulated in EG, compared with CG. It is concluded that after exercise induced cardiac remodeling by endurance training, CGRP synthesis, reserve, release is enhanced but gene expression down-regulated. With exhaustive exercises, exercise induced hypertrophied heart keeps physiological level of CGRP gene expression and elevated release.
