摘 要:目的:研究丹参酚酸A(salvianolic acid A,SalA)对大鼠局灶性脑缺血损伤的保护作用及特点。方法:利用线栓法制作大鼠左侧大脑中动脉阻塞(MCAO)模型,于苏醒时和术后24h进行神经行为学评分;24h评分后断头取脑测定脑梗塞指数、脑指数及脑含水量;应用HE染色观察SalA干预后缺血区病理学改变。结果:SalA可以改善MCAO大鼠神经功能缺损状态,缩小梗塞面积,减轻脑水肿,降低相关脑区神经元损伤程度。综合分析SalA的作用体现出比丹参酚酸B(salvianolic acid B,SalB)更明显的趋势,SalA三个剂量中以2.5mg/kg组效应更强。结论:针对脑缺血损伤,SalA的保护作用有优于SalB的趋势,而本实验中表明Sa-lA的量效关系不明显。Objective: To explore the neuroprotective effects of salvianolic acid A on focal cerebral ischemia in rats. Methods: Focal cerebral ischemia was induced by permanent occlusion of the proximal portion of left middle cerebral artery occlusion(MCAO). A neurological examination was performed on each rat come round, 24 hours after ischemia by the method of Berderson and Zhang. The infracted size was measured by 2,3,5-triphenrytetrazolium chloride(TIC) staining technique at 24 hours post-ischemia. Effect of salvianolic acid A on the histological neuronal changes were observed by means of HE staining. Results : Salvianolic acid A significantly reduced infracted size, cerebral index, water content and score of neurological deficits. Protective effects of SalA on cerebral lesion were better than ones of Sal B. Conclusion : Salvianolie acid A was a good neuroprotective agent, but its dose-effect relationship was not evident.
