摘  要:目的:研究槲芪散及其君药槲寄生对人肝癌细胞生长和端粒酶活性的影响。方法:用不同浓度的槲芪散及其君药槲寄生提取物作用于人肝癌细胞(SMMC-7721)后,用四甲基偶氮唑比色法(MTT)检测SMMC-7721细胞增殖。用实时荧光定量PCR法检测SMMC-7721的端粒酶活性。结果:(1)MTT结果显示槲芪散、槲寄生多糖和总碱能抑制SMMC-7721细胞,随药物作用时间的延长作用增强。(2)槲芪散、槲寄生多糖和槲寄生总碱能明显降低SMMC-7721端粒酶的活性。结论:槲芪散及槲寄生多糖和总碱均能抑制SMMC-7721增殖,降低SMMC-7721端粒酶的活性。Objective: To study the effect on human hepatocarcinoma cells and telomerase activity induced by Huqi San (HQS) and its principal drug mistletoe extracts Methods: After the SMMC -7721 induced by different concentration, we select the MTT to detect the proliferation of SMMC-7721; use the fluorescent quantitative PCR to detect the telomerase activity of SMMC -7721. Results: (1)The results of MTT showed that compared to control group, the inhibition ratio of SMMC -7721 induced by the low -dose mid -dose and high -dose of HQS, mistletoe polysaccharides and total alkaloid of mistletoe increase gradually. (2) The low - dose mid - dose and high - dose of HQS, mistletoe polysaccharides and total alkaloid of mistletoe have the effect of degrading the telomerase activity of SMMC -7721 compared to normal control group and C - DDP ( cis - diamminedichloroplatinum) group. Conclusions: Huqi San (HQS) and its principal drug mistletoe polysaccharides and total alkaloid of mistletoe can all inhibit the proliferation of SMMC -7721 and degrade the telomerase activity of SMMC -7721.
