摘  要:目的:基于包封率优化甲氨蝶呤红细胞载体(MTX-RBC)制备工艺。方法:按正交实验L9(34)表格设计,采用高效液相色谱柱法间接测定红细胞载体的包封率,考察不同浓度MTX、全血与高渗葡萄糖比例、脱水时间和药物浸润红细胞时间等4个因素在3种不同水平条件下对包封效率的影响。结果:各水平条件下,包封率均值最高分别出现在全血与高渗葡萄糖比例为3∶2、脱水时间为40 min、MTX溶液浓度250μg.mL-1、药物浸润红细胞时间30 min。此时包封率亦最高,达41.58%。结论:确定新鲜全血以3∶2比例在高渗葡萄糖溶液中脱水40 min,再在浓度为250μg.mL-1MTX溶液中重涨30 min为最优方案。Objective: To optimize the conditions for methotrexate-red blood cell (MTX-RBC) preparation. Method: Calculated and compared the envelopment efficiencies of MTX-RBC under different conditions listed in the L9 (34) orthogonal form. Result: The highest envelopment efficiency in each level appeared when the ratio of whole blood to 50% glucose solution was 3: 2, the dehydration time was 40 min, the MTX solution concentration was 250 μg·mL^-1 , and the re-enclosed time was 40 min, respectively, could reach to 41.58%. Conclusion: The best condition for MTX-RBC preparation was the RBC in fresh whole blood dehydrating 30 min in hypertonic glucose solution with the ratio of 3: 2 and then recovering its volume in 250 μg·mL^-1 MTX solution during another 30 min.
