摘 要:运用随机扩增多态DNA(RAPD)标记对分布于我国4省6病区63份茶藨生柱锈菌寄主红松和华山松进行遗传多样性分析。用6个随机引物对寄主松针进行PCR扩增,共得到138个DNA位点,其中多态位点117个,多态位点比率为84.78%,RAPD的Shannon指数遗传多样性为0.3155,RAPD的Nei指数基因分化系数为0.5255。不同来源寄主的遗传多态性规律顺序:辽宁清原红松>云南洱源华山松>山西沁水华山松>云南巧家华山松>四川广元华山松>云南东川华山松。从RAPD聚类分析树状图可以看出,先将63个松针样品分成2个不同的RAPD组群(红松组群和华山松组群),然后又将不同地理来源的华山松组群按地理区域分成5个亚群,同一采集的华山松亚群中又分成感病组和抗病组。研究表明,种间的遗传多样性是由种的遗传属性所决定的,而地理因素只是影响种内遗传多样性的一个因子。Genetic diversity analysis of the 63 host samples of Cronartium ribicola on Korean pines and P. armandii from six infected areas of four provinces in China based on RAPD marking. Six random primers are used in PCR amplification of host needles, generating a total of 138 RAPD markers, inclu-ding 174 polymorphie loci, which is 84.78% polymorphie. Shannon index in RAPD is 0.3155, and dif-ferentiation coefficient of Nei index in RAPD is 0.5255. Polymorphie sequence of different hosts is : Ko-rea pine in Qingyuan of Liaoning 〉 P. armandii in Eryuan of Yunnan 〉 P. armandii in Qinshui of Shanxi 〉 P. armandii in Qiaojia of Yunnan 〉 P. armandii in Guangyuan of Siehuan 〉 P. armandii in Dongehuan of Yunnan. RAPD clustering analysis dendrogram shows the P. armandii samples are divided into 2 dif- ferent RAPD groups (Korean pine group and P. armandii group) , and then the P. armandii group is fur-ther divided into 5 subgroups by geographic regions, the P. armandii subgroups from the same geographic regions are divided into infected and resistant groups. This demonstrates that genetic diversity of different species is attributable to the species genetic properties, and that geographical factor is one of the factors for intra-species generic diversity.
