摘 要:目的:探讨茵陈蒿汤对ANIT急性肝损伤模型的保护作用,并初步探讨其对肝细胞摄取、转化、排泄胆红素的影响,揭示利胆退黄的作用机理,为后续实验建立中医阳黄证黄疸动物模型奠定基础。方法:采用α-异硫氰酸萘酯(ANIT)灌胃诱导大鼠急性肝损伤,观察大鼠ALP,ALT,AST,γ-GT活性及T-BIL,D-BIL,β-G和MDA含量,评价茵陈蒿汤保肝作用。结果:茵陈蒿汤低剂量组能使肝损伤生化指标明显改善,具有显著的保肝作用,而高、中剂量组保肝作用不明显。结论:茵陈蒿汤具有保肝利胆退黄的作用,其机理可能与降低肝细胞β-G、MDA含量,促进胆红素代谢,降低脂质过氧化物含量有关。Objective: To observe the protective effect and mechanism of Yin-ChenHao-Tang(YCHT) against the hepatotoxicity induced by ANIT in Wistar rats,investigate the influence of YCHT on bilirubin,reveal the mechanism of normalizing gallbladder to cure jaundiceand,and establish the foundation of jaundice model for the future experiment.Methods:Wistar rats were orally administered ANIT to induce acute hepatic injury.The protective effect of YCHT was evaluated by analyzing the activities or contents of ALP,ALT,AST,γ-GT,T-BIL,D-BIL,β-G.and MDA., and the experimental data were statistically analyzed by ONE-WAY ANOVA(SPPSS 10.0).Results:Low dose of YCHT significantly improved all the biochemical indicators at 48h after hepatotoxin treatment.However,High dose only reduced the elevation of ALP,ALT,AST,MDA and middle dose only antagonized ALP,ALT,AST,γ-GT,MDA.Both of them didn't show enough hepatoprotective effect.Conclusion:YCHT has hepatoprotective effect with dose-effect relationship and its mechanism may relate to reducing the hepatocyte content of β-G.,promoting the excretion of bilirubin and improving the metabolism of bilirubin.
