心肌联合注射纤维蛋白封闭剂在细胞移植中应用的研究

摘　　要：目的:探讨心肌注射骨髓间充质干细胞(mesenchymal stemcells,MSCs)联合应用纤维蛋白封闭剂(FibrinSealant,FS),对细胞滞留量的影响。方法:(1)提取、分离、培养、扩增小型猪骨髓MSCs备用;(2)小型猪随机分为2组,成功建立急性心肌梗塞模型,然后分为单纯MSCs移植组和MSCs联合纤维蛋白封闭剂移植组,对比移植6周后细胞的滞留量及新生血管的密度,超声学评价心脏功能。结果:6周后MSCs+FS组荧光标记的MSCs数量、新生血管密度均较MSCs组显著增多(P<0.01),MSCs+FS组左室短轴缩短率(FS%)、心肌射血分数(EF%)较MSCs组显著提高(P<0.05,<0.01)。结论:经心外膜心肌注射MSCs联合纤维蛋白封闭剂较单纯心肌注射MSCs能够明显提高移植细胞的滞留存活数量,增加新生血管密度,改善心肌收缩功能。　Objective：To investigate the effects of intramyocardial injecting bone marrow mesenchymal stem cells combined with fibrin sealant in cells transplantation.Methods：（1）Prepared for isolation,purification,culture,expansion of bone marrow MSCs.（2）Mini-swine were randomly assigned to establish models of acute myocardial infarction and then were performed by the subsequent procedure：intramyocardial MSCs injection alone（Group MSCs）and intramyocardial MSCs injection combined with FS（Group MSCs＋FS）.Six weeks postoperatively,quantitative analysis of fluorescent-labeled cells was performed by Image Pro Plus 4.5 software.Likewisely,neovascularization by histochemically staining was analyzed as well.Cardiac function was assessed by echocardiorgaphy.Results：Six weeks postoperatively,both the retention number of positively labeled cells and new vessels density in Group MSCs＋FS showed significant increase compared with those in Group MSCs（P〈0.01）.Myocardial function of EF% and FS% in Group MSCs＋FS markedly improved compared with that in Group MSCs（P〈0.05,〈0.01）.Conclusion：Intramyocardial injecting bone marrow MCSs combined with fibrin sealant can increase the retention of transplanted cells,promote neovascularization formation and enhance myocardial systolic function.
