摘  要:目的探讨日本血吸虫卵对小鼠溃疡性结肠炎保护作用的免疫调节机制。方法以10 000个日本血吸虫卵经腹腔注射免疫小鼠4次,每次间隔1周,末次免疫后6 d用三硝基苯磺酸(TNBS)诱导结肠炎模型,观察虫卵免疫组和未免疫组小鼠的细胞因子和调节性T细胞(regulatory T cells,Tregs)亚群CD4+CD25+Foxp3+水平的变化。结果未经虫卵免疫的小鼠经TNBS诱导后结肠炎症状明显,细胞因子IFN-γ水平高达3.47±0.87 ng/ml,IL-4I、L-5和IL-10水平则较低分别为146.06±45.76 pg/ml,140.32±39.01 pg/ml,385.91±134.89 pg/ml;经虫卵免疫的结肠炎小鼠的炎症得到明显控制,IFN-γ表达被明显抑制其水平为1.53±0.51 ng/ml,IL-4I、L-5和IL-10水平显著升高分别为598.50±135.90 pg/ml,217.86±77.82 pg/ml,799.651±197.93 pg/ml。经虫卵免疫的结肠炎小鼠脾脏CD4+CD25+FoxP3+T细胞亚群比例为(3.55±0.81)%,显著高于未免疫组结肠炎小鼠的(2.31±0.39)%。结论日本血吸虫卵免疫对TNBS诱导的结肠炎小鼠具有保护作用,其保护性机制可能与CD4+CD25+FoxP3+T细胞对Th1过度反应的反馈性调节有关。Objective To study the immune modulatory mechanisim of the protection of mice against colitis induced by trinitro-benzene-sulfonic acid(TNBS) with Schistosorna japonicum eggs. Methods Mice were immunized 4 times with 10 000 schistosome eggs by intraperitoneal injection with one-week interval. On day 6 after the last immunization the colitis was induced by TNBS and pathologic changes on colon, levels of cytokines and CD4^+CD25^+FoxP3^+ T cells of mice were observed and detected in groups of egg-immunized and unimmunized colitis mice. Results In egg-unimmunized colitis mice, after TNBS induction, the inflammation in colon was obvious. And it was characterized with high Thl cytokine IFN-γ level (3.47±0.87 ng/ml) and lower Th2 cytokine IL-4, IL-5 and IL-10 level (146.06±45.76 pg/ml, 140.32±39.01 pg/ml, 385.91±134.89 pg/ml, respectively). While in egg-immunized colitis mice, the inflammation was suppressed greatly. And schistosome eggs exposure significantly diminished the expression of IFN-γ (1. 53 ±0.51 ng/ml) and enhanced the production of IL-4, IL-5 and IL-10 (598.50± 135.90 pg/ml, 217.86± 77.82 pg/ml, 799. 651± 197.93 pg/ml, respectively) in colitis mice. The proportion of CD4^+ CD25^+ FoxP3^+ T ceils in splenocytes was (3.55±0.81) % and higher than that in TNBS-induced colitis mice alone (2.31±0.39) %. Conclusion The eggs of S. japonicurn can inhibit the development of colitis induced by TNBS in mice and the modulation of CD4^+CD25^+FoxP3^+ T cells to the over-expressed Thl immune response may be involved in the mechanism.
