摘 要:目的考察超临界CO2流体萃取伸筋草有效成分的最佳工艺,进一步验证萃取物的药效。方法采用正交实验设计,以萃取物得率为指标,考察萃取压力、萃取温度、解析压力及解析温度4个因素的影响;采用热板法、醋酸扭体法、鼠耳二甲苯致炎法、棉球肉芽肿致炎法观察萃取物的镇痛抗炎效果。结果最佳萃取工艺条件为萃取压力20 mPa,萃取温度35℃,解析压力8 mPa,解析温度55℃,萃取时间4 h;萃取物对热致痛和冰乙酸所致的小鼠扭体疼痛具有良好的镇痛作用,对二甲苯致小鼠耳炎有显著的消炎作用。结论超临界CO2流体萃取技术用于伸筋草有效成分提取,方法简单,适用于工业生产。伸筋草超临界萃取物具有显著镇痛抗炎的药理作用。Objective To study the optimal technology of supercritical-CO2 fluid extraction( SFE-CO2 ) on Lycopodium japonicum Thunb. and validate the drug action of extract. Methods The yield of extractant was used as index to evaluate the technologies based on orthogonal design , in which 4 factors considered were extraction temperature and pressure , separation temperature and pressure ;the anti - inflammation and analgesia pharmacological actions was observed by drug action experiment. Results The optimum extraction conditions were : extraction pressure 20MPa, temperature 35℃ ;separation pressure 8MPa, temperature 55℃ ; extraction time 4 hours. The extract had good analgesia pharmacological action to pain caused by heat in mice and by acetic acid causing body distortion test in mice. It also had significant depressions to the inflammation caused by dimethyl benzene on the ear of mice. Conclusion Supercritlcal fluids extraction method is simple for the activities of Lycopodium japonicum Thunb. , and useful for the large scale production. The extract has significant anti-inflammation and analgesia pharmacological actions.
