摘  要:目的:探讨人肝细胞生长因子(hHGF)基因肌肉电转染对大鼠肾脏缺血再灌注损伤的保护作用。方法:♂SD大鼠20只,随机分为2组,即研究组和对照组,每组10只。研究组大鼠接受hHGF肌肉电转染,在热缺血前3d将含有hHGF基因的质粒用电转染技术转入大鼠双侧胫前肌肉内,3d后建立大鼠肾热缺血模型,测定hHGF的血药浓度,并对缺血再灌注肾脏进行功能和组织学评价。结果:研究组肾组织及血浆hHGF基因表达水平显著高于对照组。研究组血肌酐水平较对照组低。组织学评价结果显示,研究组肾小管细胞凋亡评分显著低于对照组。研究组淋巴细胞、巨噬细胞及中性粒细胞浸润均较对照组少。结论:本研究结果表明hHGF基因肌肉电转染对大鼠肾脏缺血再灌注损伤具有保护作用,并提示肌肉hHGF基因电转染有可能成为防治移植肾缺血再灌注损伤的安全有效的方法之一。OBJECTTVE To investigate the efficacy of muscle electrotransfer of hHGF gene in reducing renal ischemia-reperfusion injury in rats. METHODS 20 male SD rats were randomly divided into two groups, 10 in study group and another 10 in control group, hHGF gene was transferred into both sides of tibial muscles of the rats in study group by means of electrotransfer assay 3 days before renal warm ischemia injury. The concentration of hHGF were determined, the renal function and histological change of the kidney after ischemia-reperfusion injury were assessed. RESULTS Higher expression levels of hHGF gene in the plasma and renal tissue were observed in study group when compared to that of controls. The serum creatinine levels in study group were significantly lower than controls and the rats in study group had rapider recovery of renal function when compared to controls. It was shown by histological evaluation that there were less apoptosis score of tubular cells and less lym phocyte, macrophage and neutrophil infiltration in study group, compared to controls. CONCLUSION It is indicated that the muscle electrotransfer of hHGF gene was effective in preventing renal ischemia-reperfusion injury. Furthermore it is suggested that the muscle electrotransfer of the gene before renal transplantation might be effective in reducing the ischemia-reperfusion of the transplanted kidney.
