氯沙坦对兔颈动脉球囊损伤后血IL-1β和IL-10的影响

摘　　要：目的探讨血管紧张素Ⅱ1型受体(AT1)和拮抗剂(ARB)对抑制内膜增生的可能机制。方法12只新西兰白兔随机分为实验组和对照组,每组6只。实验组给予氯沙坦处理,采用酶联免疫吸附法(ELISA)和放射免疫法检测两组颈动脉球囊损伤后的血清白细胞介素(IL-1β、IL-10)水平,并比较相关关系。结果血清IL-1β水平两组均于术后6h开始升高,48h达高峰,随后逐渐降至基线水平;但在6、24、48h及1周时对照组显著高于实验组。血清IL-10水平在整个实验过程中实验组均显著高于对照组(P均<0.01)。血清IL-1β水平与IL-10水平无论在实验组还是对照组均无相关关系(r=-0.15,P=0.37;r=-0.28,P=0.10)。结论在动脉损伤后氯沙坦可以通过降低IL-1β和升高IL-10的血清水平来发挥抗炎作用,这可能是ARB抑制内膜增生的机制之一。Objective To investigate the possible mechanisms of angiotensin Ⅱtype 1 （AT1） receptor blecker （ARB） in inhibiting neointimal hyperplasia. Methods Twelve New Zealand white rabbits were randomly divided into experimental group and control group, each group had six rabbits. From seven days before to 2 weeks after carotid artery injury , losartan was administered in the experimental group by intubation feeding whereas pure water was administered by the same method in the control group. Serum concentrations of IL - 1β IL - 10 were measured in beth groups by enzyme - linked immunosorbent assay （ELISA） and radioimmunoassay（RIA） immediately or 6,24,48 hours and 1,2 weeks after carotid artery injury. The relation between IL - 1β and IL- 10 was compared. Results Serum concentrations of IL- 1β in beth groups elevated 6 hours after angioplasty, peaked at 48 hours, and gradually decreased to baseline thereafter. But the concentrations of IL - 1β in the experimental group were significandy decreased at 6,24,48h and 1 wk than those in the control group（0.050 ± 0.022 ng/ml vs 0.018 ± 0.014 ng/ml,0.070 ± 0.020 ng/ml vs 0.034 ± 0.014 ng/ml,0.105 ± 0.012 ng/ml vs 0.058 ± 0.012 ng/ml,0.026 ± 0.007 ng/ml vs 0.015 ± 0.006 ng/ml, P 〈 0.05, respectively）. Serum concentrations of IL- 10 were significantly higher in the experimental group throughout the experiment in comparison with the control group （ P 〈 0.05 ）. There was no positive or negative correlation between the serum concentrations of IL - 1β and those of IL- 10 either in the experimental group or in the control group （ r = - 0.15, P = 0.37 ; r = - 0.28, P = 0.10）. Conclusion In this study, losartan could decrease the serum concentrations of IL - 1β and increase those of IL - 10 after carotid artery injury, this anti - inflammatory, effect may play an important role in the mechanisms that ARB inhibits intimal hyperplasia .
