拟南芥胚发育的整体透明观察

摘　　要：以受精后不同发育时期的拟南芥胚珠为材料，经固定、脱水，用水杨酸甲酯整体透明，在微分干涉衬显微镜下清晰地观察到胚发育的过程。A whole clearing technique has been developed forembryological observations on whole mounted spcimens.Ovules of Arabidopsis thaliana were fixed for a proper time, followed by steps of dehydration,cleared in methyl salicylate,and finally viewed by differential interference contrast microscope (DIC), the embryos were clearly visible. The special significance of this technique for in vitro embryological studies of higher plants is emphasized.
