摘 要:目的探讨临床分离白念珠菌氟康唑耐药株/敏感株多药耐药蛋白基因CDR1、CDR2、MDR1的表达情况。方法试剂盒法抽提白念珠菌氟康唑耐药株/敏感株的总RNA后,逆转录成cDNA,采用实时定量PCR技术观察白念珠菌多药耐药蛋白基因的表达。结果白念珠菌耐药株组CDR2基因的ΔCT值为7.52±2.53,敏感株组为9.28±3.15,两组比较,t=2.37,P<0.05,耐药株的表达量要高于敏感株。结论白念珠菌CDR2基因的高表达与氟康唑耐药有关。Objective To explore the relationship between fluconazole resistance and expression of multidrug resistant protein genes, including CDRI, CDR2, MDRI. Methods The total RNA was extracted from fluconazole-resistant and -susceptible Candida albicans isolates, and cDNA was synthesized. The expression of CDRI, CDR2 and MDRI genes was then detected by quantitative real-time PCR. The ACT ( threshold cycle ) value was obtained by subtracting the CT value of 18S rRNA from that of the targeted gene. Results The ACT value of CDR2 was significantly lower in fluconazole-resistant isolates than in fluconazolesusceptible isolates ( 7.52 ±2.53 vs. 9.28 ± 3.15, t = 2.367, P 〈 0.05 ), indicating the expression of CDR2 gene was higher in the fluconazole-resistant isolates than in the fluconazole-susceptible isolates. Conclusion It is likely that the over-expression of CDR2 gene is relevant to fluconazole resistance in Candida albicans.
