摘  要:目的:研究米非司酮和孕三烯酮对体外培养的异位的和正常的子宫内膜间质细胞生长增殖的影响。方法:用四甲基偶氮唑蓝(MTT)比色法检测米非司酮或孕三烯酮处理异位子宫内膜间质细胞0-60小时细胞抑制情况,并绘制细胞生长曲线;采用流式细胞仪检测药物作用36小时异位的和正常的子宫内膜间质细胞的细胞周期及凋亡率变化。结果:米非司酮和孕三烯酮呈时间和剂量方式抑制细胞的生长增殖,且相同浓度后者作用更强。药物作用36小时,各组细胞均发生从G1期到S期的抑制,且各组细胞凋亡率明显升高。结论:①米非司酮和孕三烯酮均抑制细胞增殖,诱导细胞凋亡,但后者作用更强。②米非司酮和孕三烯酮对正常子宫内膜间质细胞的抑制作用明显不如对异位细胞的作用强。Objective:To investigate mifepristone and gestrinone's effects on growth and proliferation in isolated ectopic and normal endometrial stroma cells in vitro. Methods: Endornetrial stroma cells were cultured and exposed to mifepristone or gestrinone of different doses. The inhibition of the cells during 60 hours was determined by melhylthiazolyl tetrazolium (MTT) assay, and the cell growth curve was made. At 36 hours the cell cycle and apoptosis rate were monitored by flow cytometry (FCM). Results: Both mifepristone and gestrinone could inhibit the growth and proliferation of endometrial stroma cells in a dose - and time - dependent manner. After 36 hours, the cell cycles were inhibited from G1 stage to S stage, with increased apoptotic rate. Conclusions:①Mifepristone and gestrinone can both significantly inhibit proliferation and induce apoptosis. When compared at the same concentration, the latter has stronger effect. ②The inhibition effect of mifepristone and gestrinone to the ectopic endometrial stroma cells was stronger than to those from normal endometrium.
