免疫刺激DNA序列与尘螨过敏原联用治疗变应性鼻炎的实验研究

摘  要:目的:观察免疫刺激DNA序列(ISS)与尘螨过敏原(Df)合用,对Df致敏的变应性鼻炎(AR)大鼠模型鼻腔过敏性炎症的作用及其维持时间。方法:SD大鼠32只,分ISS组(A组)、ISS加Df组(B组)、Df组(C组)和正常对照组(D组),每组各8只。A、B、C组用Df致敏及激发。各组在第1次激发后6周中,每周采血1次测特异性IgE,最后处死大鼠,组织病理学方法观察鼻黏膜改变,ELISA测血清IL-4、IL-5、IFN-γ和IL-12水平。结果:A、B组IFNγ-和IL-12水平明显高于C组(P<0.01),而A、B组IL-4和IL-5水平明显低于C组(P<0.01)。血清特异性IgE前5周B组与C组比较差异有统计学意义(P<0.01),第6周B组Df特异性IgE水平有回升,但与C组比较差异仍有统计学意义(P<0.05);而A组与C组比较差异无统计学意义(P>0.05)。结论:ISS与Df合用对Df致敏的AR大鼠模型鼻腔过敏性炎症有很好的抑制作用,其抑制作用可维持6周。Objective:To observe the suppressive effects of immunostimulatory DNA seqences(ISS) in conjunction with dermato phagoides farinae allergen(Df ) on nasal cavity inflammation in rats of experimental allergic rhinitis (AR). Method:Thirty-two rats were divided into 4groups: group ISS(A), group ISS+ Df (B), group Df (C) and group normal saline(D). Rats in groups A,B and C were sensitized and challenged with Df. Blood samples were obtained every week for six weeks, Df specific IgE was measured by ELISA. The nasal mucosa were studied patho.logically. The levels of serum interferon γ(IFN-γ),IL-12, IL-4 and IL-5 were determined by ELISA. Result: The serum level of slgE in group B was significantly lower than that in group C in six weeks, but that in group A was not significantly different to that in group C. The mean levels of serum IFN-γ and IL- 12 in A, B group were significantly higher than in C group(P〈0.01). But IL-4 and IL-5 level was much lower (P〈0.01). Conclusion:ISS+ Df have inhibited effectiveness on allergic nasal cavity inflammation in rats and its action time is 6 weeks.
