摘 要:目的:探讨2002—2004年天津市流行的麻疹病毒株基因型别。方法:采用B95a细胞培养法分离麻疹病毒(MV),用逆转录-聚合酶链反应(RT-PCR)方法扩增分离出的MVN基因C端片段,并进行序列分析确定我市流行的MV型别,构建种系发育树,进行遗传距离、基因变异分析。结果:天津市2002—2004年分离到的16株MV经RT-PCR和序列分析后显示均为H1基因型,亚型分析1株为H1b亚型,15株为H1a亚型。结论:我市分离出的MV均为H1型,其中又以H1a占绝对优势,提示H1a型为我市的优势流行株。Objective: To study the genotype of the measles viruses circulated in Tianjin from 2002 to 2004. Mothods: Measles viruses were isolated by B95a cell line and identified through RT-PCR. The C-terminal region of the N-gene was amplified and sequenced. Phylogenetic analysis and the diversity analysis were performed. Rosults: Sixteen strains of measles viruses were isolated and they were all H1 genotype by sequence analysis including 1 Hlb and 15 Hla. Conclusion: All measles viruses isolated in Tianjin from 2002 to 2004 included H1 genotype and Hla is the mainly predominant subgenotype. It suggests that the H 1 a genotype is the predominate virus in Tianjin.
