摘 要:目的:研究犬脑枪弹伤后心肌细胞凋亡及bcl-2的表达.方法:建立犬颅脑枪弹伤模型,制备心肌组织切片.采用末端脱氧核苷酸转移酶介导的生物素脱氧尿嘧啶核苷酸缺口末端标记法(TUNEL)检测心肌细胞凋亡以及用免疫组织化学方法检测bcl-2的表达.结果:对照组偶见心肌细胞凋亡及bcl-2的表达,枪弹伤后3h凋亡心肌细胞及bcl-2表达明显(P<0.01).心室壁内侧凋亡细胞分布基本一致(P>0.05),内外侧壁凋亡细胞分布不同(P<0.01).心室壁内侧以及内外侧壁心肌细胞bcl-2表达差异无统计学意义(P>0.05).结论:犬脑枪弹伤后可造成心肌细胞损害和bcl-2表达.AIM: To study the apoptosis of and bcl-2 expression in eardiomyocytes following eraniocerebral gunshot wounds in dogs. METHODS: Experimental craniocerebral gunshot wound models were established in mongrel dogs. Then the samples of cardiac muscle at different areas were taken from the 5 gunshot wounded dogs and 5 normal control dogs. Apoptotie eardiomyo- eytes were detected by TUNEL, and Bel-2 positive eardiomyocytes were observed by immunohistochemieal method. RESULTS: Apoptosis and bel-2 expression were seldomly seen in the control eardiomyocytes. Apoptosis and bel-2 positive expression in eardio- myocytes was siguifieanfly increased at 3 h after eraniocerebral gunshot wounds ( P 〈0.01 ). The apoptotie eardiomyocytes in inside ventricle wall was similar ( P 〉 0.05 ) , but the localization of apoptotic eardiomyocytes was different between outside ventrieular wall and inside ventrieular wall ( P 〈 0.01 ). The expressions of bel-2 in different areas showed no difference (P 〉 0.05 ). CONCLUSION: The eraniocerebral gunshot wounds in dogs may lead to bel-2 positive expression and the damage of cardiac muscle.
