摘  要:目的:制定美洲大蠊药材的鉴别、检查及含量测定方法。方法:采用薄层色谱法定性鉴别,色谱条件为正丁醇∶乙酸∶水(4∶1∶1),0.5%茚三酮显色。分光光度法进行总氨基酸的含量测定,茚三酮显色后检测波长为570nm。采用水分测定法检查水分的最低控制限为12%。采用热浸法检查乙醇浸出物的最低控制限为10%。结果:薄层色谱鉴别斑点清晰,易于识别,分光光度法测定总氨基酸含量,回归方程:A=0.067943C+0.009143,r=0.9999,酪氨酸含量在1.0~10.0μg范围内呈良好的线性关系。结论:该方法简便易行,重现性好,可用于美洲大蠊药材的质量控制。Objective: To study the methods of identification and assay in Periplanta americana. Methods: TLC was used and the chromatographic condition was as follows: n-Butanol, Aceticacid and water (4:1:1); when determining the content of total amino acids by pectrophotometry after ninhydrin colouring, the determination wavelength was 570nm in ninhydrin colouring. The minimum control limit of the water content was 12% by using water content assay method, The content of ethanol soluble extraction should be no less than 10% by hot dip, Result: The method of TLC was exclusive and spots were clear, the determination of amino acid by spectrophotometry, regression equation is A=0.067943C+0.009143, r=0.9999, Tyrosine content varied from 1.0 to 10.0 μg and the linearity is good, Conclusion: The methods are simple and accurate. The method can be used for the quality control of Periplanta americana.
