针刺手厥阴经穴对缺血再灌注损伤心肌细胞蛋白激酶C表达的影响

摘　　要：目的研究针刺手厥阴经穴对缺血再灌注损伤过程中心肌细胞蛋白激酶C(PKC)表达的影响,探讨PKC介导的信号转导机制在心肌缺血再灌注损伤中的作用。方法电针大鼠手厥阴经穴位20分钟后,结扎左冠状动脉前降支40分钟,心电图监测,再电针穴位20分钟,松扎,恢复灌流60分钟,摘取心脏,免疫组化方法分析细胞蛋白激酶C的表达。结果缺血再灌注模型组PKC表达的阳性率明显增高,针刺手厥阴经穴后阳性表达率明显降低(P<0.01)。结论针刺手厥阴经穴对PKC表达有明显的抑制作用,从而发挥对心肌细胞的保护作用。Objective To study the effect of needling at points of Pericardium Meridian of Hand Jueyin on the expression of protein kinase C （PKC） and signal transduced mechanism mediated by PKC in cardiocyte of ischemical reperfusion injury. Methods Points of Pericardium Meridian of Hand Jueyin of rats were given electro-acupuncture for 20 minutes. Ramus descendens anterior arteriae coronariae sinistrae was ligated for 40 minutes with electrocardiogram monitoring. Ligation ceased after another 20 minutes of electroacupuncture. Heart was extracted after 60 minutes of reperfusion. The expression level of PKC was tested by immunohistochemical method. Results The expression level of PKC increased in model group but decreased significantly after acupuncture （P〈0.01）. Conclusion Needling points of Pericardium Meridian of Hand Jueyin can obviously inhibit the expression level of PKC and protect cardiocyte.
