心衰康颗粒对“心梗”后大鼠心肌Ⅰ、Ⅲ型胶原mRNA表达影响的研究

摘　　要：目的:观察心衰康颗粒对急性心梗后大鼠心肌Ⅰ、Ⅲ型胶原mRNA表达的影响,阐释心衰康颗粒干预左室重构的作用机理。方法:采用结扎Wistar大鼠左冠状动脉根部建立心梗后左室重构动物模型,随机分为正常组、假手术组、模型组、心衰康大剂量组、心衰康小剂量组和卡托普利对照组,分别药物干预2月后,观察大鼠心肌Ⅰ、Ⅲ胶原mRNA表达的变化。结果:①模型组Ⅰ、Ⅲ型胶原mRNA表达明显高于正常组及假手术组(P<0.05)。②与模型组比较,所有给药组I、Ⅲ型胶原mRNA表达明显下降(P<0.05),大剂量组与西药组相比无统计学意义(P>0.05),与小剂量组比较有显著差异(P<0.05)。③小剂量组与西药组比较无统计学意义(P>0.05)。结论:心衰康大剂量组能够抑制心肌间质Ⅰ、Ⅲ型胶原蛋白的表达,此应为干预实验性心梗后大鼠左室重构的机制之一。Objective： To observe the influence of Xinshuaikang Granula on the expression of collagen Ⅰ , Ⅲ mRNA of AMI （acute myocardial infarction） rats, and to expound the effective mechanism of left ventricle reconstitution intervened by Xinshuaikang Granula. Methods： The left ventricle reconstitution model rats were built by ligating the root of left coronary artery of Wistar rats and randomly divided into six groups （normal group, sham operation group, model group, large dosage group of Xinshuaikang Granula, small dosage group of Xinshuaikang Granula and Captopril control group）. After 2 months respectively intervened by drugs, the change of the expression of collagen Ⅰ , Ⅲ mRNA was observed. Results： 1. The expression of collagen Ⅰ , Ⅲ mRNA of model group was apparently higher than that of normal group and sham operation group（ P 〈 0.05 ）. 2. Compared with that of model group, the expression of collagen Ⅰ , Ⅲ mRNA of all groups treated by drugs decreased evidently （P〈0.05）. 3. When large dosage group was compared with the Western medicine group, there had no statistical significance （P 〉 0.05）. Conclusion： Large dosage group of Xinshuaikang Granula can control the expression of collagen Ⅰ , Ⅲ mRNA cardiac muscle interstitial, and it should be one of the left ventricle reconstitution mechanisms of experimental rats＇ AMI.
