摘 要:目的探讨锌对宫内外铅暴露幼鼠微量元素铁、血红蛋白含量及学习记忆有无保护作用。方法将16只受孕Wistar健康大鼠分为4组,即正常对照组(A组)、高铅组(B组)、高锌+高铅组(C组)、低锌+高铅组(D组)。从受孕第14天起分别给予相应饲料及饮用水,子代断乳后饲以与其母亲相同的饲料和饮用水。于生后8周测体质量,训练主动回避反应习得率、保持率,测全血及海马铅、铁、锌含量和血红蛋白含量。结果①全血及海马的铅含量:B、D组均显著高于A、C组,C组与A组间无显著差异。②全血及海马的锌含量:C组与A组相近,B、D组均显著低于A、C组。③全血铁含量:C组虽略低于A组,但无显著差异;B组显著低于A组;D组则显著低于A、C组。海马铁含量:C组与A组无显著差异,但B、D组均显著低于上述2组,以D组更甚。④血红蛋白含量:B、C组与A组间无显著差异,D组显著低于上述3组。⑤AAR习得率与保持率:C组与A组相近,其余各组均显著低于A、C组,其中又以D组最明显。结论亚临床铅中毒时,全血铁及海马铁、血红蛋白含量显著低于正常对照组,其中又以低锌高铅组更明显。适当增加饲料锌含量可改善机体铁营养状况。Objective To study the effect of endouterine and extra-aterine exposure to lead on iron concentrations in the blood and hippocampus, hemogiobin level and the learning memory in young rats and the protective effect of zinc in this event. Methods Sixteen pregnant Wistar rats were randomized into 4 groups, group A fed with diet containing Zn 17.5 mg/kg and normal water, group B with Zn 17.5 mg/kg and water with PbAC · 3H2O 500 mg/L, group C with Zn 168.7 mg/kg and PbAC · 3H2O 500 mg/L, and group D with Zn 〈5 mg/kg and PbAC · 3H2O 500 mg/L from the 14th day after gestation. After weaning, the pups were fed likewise for 8 weeks, and then, the following procedures were manipulated in the offspring: (1)Active avoidance response (AAR) was tested in one-way shuttle box; (2)The concentrations of lead, zinc, iron in the blood and hippocampus and hemoglobin level were measured. Results (1)The lead content in the blood and hippocampus was significantly higher in group B and D than in group A and C, and the later 2 groups had no difference. (2)The blood and hippocampal zinc contents was significantly lower in group B and D than in group A and C, and the later 2 groups had similar contents. (3)The blood iron content was a little bit lower in group C than group A, significantly lower in group B than group A, and group D than group A and C. While in the hippocampus, no significantly difference was seen in group C and A, but they were higher than group B and D, especially in the later. (4)No significant difference was observed in the hemoglobin level in group C and B with group A, but the level of group D was obviously lower than in these 3 groups. (5)The acquisition and retention rates in AAR were approximative in group A and C, significantly higher than in group B and D. Conclusion In Subclinical lead poisoning, iron and hemoglobin contents in the blood and hippocampus are significantly lower than in normal control, especially in group with low zinc and high lead diet.
