摘  要:目的研究单核细胞趋化蛋白-1(monocyte chemotactic protein-1,MCP-1)对人脐静脉内皮细胞(human um-bilical vein endothelial cells,hUVEC)凋亡的影响。方法培养并鉴定人脐静脉内皮细胞;用不同浓度MCP-1(0.1、1.0、10、100μg/L)对其作用24、48h;先用MTT比色法观察细胞存活率、流式细胞技术检测细胞DNA含量及细胞周期观察其对hUVEC凋亡的影响。结果单核细胞趋化蛋白-1能诱导人脐静脉内皮细胞的凋亡(P<0.05),其效应随浓度和时间的增加而增强:MTT明显减低;细胞DNA含量减少,出现明显亚G0/G1峰(凋亡细胞峰)。结论单核细胞趋化蛋白-1能诱导人脐静脉内皮细胞凋亡。Objective To study the effects of monocyte human umbilical vein endothelial cells (hUVEC). Methods chemotactic protein-1 (MCP-1) on the growth of hUVEC were cultured in vitro and identified hUVEC that grew vigorously were stimulated for 24 h or 48 h respectively with MCP-1 (0.1, 1.0, 10, and 100 Ixg/L). The survival rate of hUVEC was first detected by MTF assay. The cell cycle and DNA content were detected and analyzed by flow cytometry. Results hUVEC were isolated from human umbilical veins and cultured, and then identified by immunofluorescence and immunohistochemistry with factor VIH and KDR. MCP-1 induced the apoptosis of hUVEC in a dose-dependentmanner and a time-dependentmanner (P 〈 0.05). Compared with control group, MTF and chromosomal DNA content decreased significantly. Conclusion MCP-1 can induce the apoptosis of hUVEC.
